The
lipid
nanoparticle
(LNP)
has
been
so
far
proven
as
a
strongly
effective
delivery
system
for
mRNA
and
siRNA.
However,
the
mechanisms
of
LNP's
distribution,
metabolism,
elimination
are
complicated,
while
transportation
pharmacokinetics
(PK)
LNP
just
sparsely
investigated
simply
described.
This
study
aimed
to
build
model
RNA-LNP
in
Hela
cells,
rats,
mice,
humans
by
physiologically
based
pharmacokinetic
(PBPK)
quantum
mechanics
(QM)
models
with
integrated
multi-source
data.
LNPs
different
ionizable
lipids,
particle
sizes,
doses
were
modeled
compared
recognizing
their
critical
parameters
dominating
PK.
Some
interesting
results
found
models.
For
example,
metabolism
lipids
was
first
limited
disassembly
rate
instead
hydrolyzation
lipids;
ability

from

Physiologically
based
pharmacokinetic
(PBPK)
modeling,

cornerstone

model‐informed
drug
development
and
precision
dosing,

parameters.

PBPK
modeling
has
advanced
globally
since
1970s,
China's
adoption

review
provides
comprehensive
analysis

to
addressing
knowledge
gaps
publication
trends,
application

preferences.

revealed
that

interaction

constitute
