The lipid nanoparticle (LNP) has been so far proven as a strongly effective delivery system for mRNA and siRNA. However, the mechanisms of LNP's distribution, metabolism, elimination are complicated, while transportation pharmacokinetics (PK) LNP just sparsely investigated simply described. This study aimed to build model RNA-LNP in Hela cells, rats, mice, humans by physiologically based pharmacokinetic (PBPK) quantum mechanics (QM) models with integrated multi-source data. LNPs different ionizable lipids, particle sizes, doses were modeled compared recognizing their critical parameters dominating PK. Some interesting results found models. ABSTRACT Physiologically based pharmacokinetic (PBPK) modeling, a cornerstone of model‐informed drug development and precision dosing, simulates disposition in the human body by integrating physiological, biochemical, physicochemical parameters. While PBPK modeling has advanced globally since 1970s, China's adoption this technology followed distinctive path, characterized accelerated growth over past 2 decades. This review provides comprehensive analysis contributions to addressing knowledge gaps publication trends, application domains, platform preferences. A systematic literature search yielded 266 original research articles from PubMed up August 08, 2024. The revealed that drug–drug interaction studies constitute largest proportion analyses China. A spiral microfluidic chip (SMC) and multi-spiral (MSMC) for lipid nanoparticle (LNP) production were fabricated using a CO2 laser engraving method, perfluoropolyether (PFPE) poly(ethylene glycol) diacrylate as photopolymerizable base materials. The SMC includes microchannel that enables rapid fluid mixing, thereby facilitating the of small uniform LNPs with size 72.82 ± 24.14 nm PDI 0.111 0.011. MSMC integrates multiple parallel structures, which high-throughput LNP without compromising quality achieves maximum capacity 960 mL per hour. fabrication technology has potential applications in pharmaceutical field due to ease fabrication, simplicity cost-effectiveness process, ability produce high-quality LNPs.
