Over the past two decades, iron-based metal-organic frameworks (Fe-MOFs) have attracted significant research interest in biomedicine due to their low toxicity, tunable degradability, substantial drug loading capacity, versatile structures, and multimodal functionalities. Despite great potential, transition of Fe-MOFs-based composites from laboratory clinical products remains challenging. This review evaluates key properties that distinguish Fe-MOFs other MOFs highlights recent advances synthesis routes, surface engineering, shaping technologies. Extremely short half-life therapeutic molecule nitric oxide (NO) plays significant roles in the functioning of various physiological and pathological processes human body, whereas doxorubicin hydrochloride (DOX) is a clinically important anticancer drug widely used cancer chemotherapy. Thus, intracellular delivery these molecules tremendously to achieve their full potential. Herein, we report novel approach for development highly water-dispersible magnetic nanocarriers codelivery NO DOX. Primarily, bifunctional nanoparticles enriched with carboxyl thiol groups were prepared by introducing cysteine onto surface citrate-functionalized Fe3O4 nanoparticles. DOX was electrostatically conjugated via moieties, group further nitrosated provide NO-releasing molecules. The developed nanocarrier exhibited good aqueous colloidal stability, protein resistance behavior, high encapsulation efficacy (65.5%) (85%), as well sustained release characteristics. Moreover, they showed superior cytotoxicity toward (A549 MCF-7) cells apoptosis induction over normal (WI26VA4) cells.
