Long noncoding RNAs (lncRNAs) are RNA molecules of 200 nucleotides or more in length that not translated into proteins. Their expression is tissue-specific, with the vast majority involved regulation cellular processes and functions. Many human diseases, including cancer, have been shown to be associated deregulated lncRNAs, rendering them potential therapeutic targets biomarkers for differential diagnosis. The lncRNAs nervous system varies different cell types, implicated mechanisms neurons glia, effects on development functioning brain. Protein aggregation is classically considered the main cause of neuronal death in neurodegenerative diseases (NDDs). However, increasing evidence suggests that alteration RNA metabolism a key factor etiopathogenesis these complex disorders. Non-coding RNAs are major contributor to human transcriptome and particularly abundant central nervous system, where they have been proposed be involved onset development NDDs. Interestingly, some ncRNAs (such as lncRNAs, circRNAs pseudogenes) share common functionality their ability regulate gene expression by modulating miRNAs phenomenon known competing endogenous mechanism. Multiple sclerosis (MS) is a chronic disease and one of the leading causes disability in young adults. The current study aims to investigate pathogenesis MS via studying regulatory role novel lncRNA MAGI2-AS3 miR-374b-5p their downstream targets PTEN/AKT/IRF-3/IFN-β relationship this pathway with severity. Moreover, it assess MAGI2-AS3/miR-374b-5p as diagnostic and/or prognostic biomarkers for MS. Overall, 150 contributors were recruited: 100 patients 50 healthy volunteers. Gene expression MAGI2-AS3, miR-374b-5p, PTEN, AKT, IRF-3 assessed using RT-qPCR, IFN-β was measured by ELISA. Compared control group, serum PTEN downregulated patients, whereas PI3K, IRF-3, upregulated patients. Furthermore, downregulated, while an expanded status scale (EDSS) ≥3.5, compared EDSS <3.5. Receiver-operating-characteristic curve analysis revealed that can be used diagnosis Remarkably, multivariate logistic AKT act independent variables directly correlated inversely EDSS. Abstract Neurological disorders such as stroke, multiple sclerosis, well the neurodegenerative diseases Parkinson's or Alzheimer's disease are accompanied even powered by danger associated molecular patterns (DAMPs), defined endogenous molecules released from stressed damaged tissue. Besides protein-related DAMPs “alarmins”, numerous nucleic acid exist in body fluids, cell-free nuclear and mitochondrial DNA different species of extracellular RNA, collectively termed self-extracellular acids (SENAs). Among these, microRNA, long non-coding RNAs, circular RNAs ribosomal RNA constitute majority RNA-based DAMPs. Upon tissue injury, necrosis apoptosis, SENAs neuronal, immune other cells predominantly association with vesicles may be translocated to target where they can induce intracellular regulatory pathways gene transcription translation. The SENA-induced signaling reactions brain appear related neuroinflammatory processes, often causally onset progression respective disease. In this review, impact diverse types on will discussed. Based accumulating knowledge field, several specific antagonistic approaches presented that could serve therapeutic interventions lower pathological outcome indicated disorders.

Cerebral ischemic stroke (CIS) is one of the primary causes death worldwide and a major cause long-term disability. Long noncoding RNAs (lncRNAs) have emerged as crucial mediators in pathology CIS; however, their potential importance yet to be discovered. Herein, we examined association four single-nucleotide polymorphisms (SNPs) with risk CIS, correlation lncRNAs, MALAT1 ANRIL, expression, serum ANRIL biomarkers for CIS. A total 100 CIS patients healthy controls were recruited study. Genotyping expression analysis SNPs carried out by qPCR. Immune cell function is critically dependent on precise control over transcriptional output from the genome. In this respect, integration of environmental signals that regulate gene expression, specifically by transcription factors, enhancer DNA elements, genome topography and non-coding RNAs (ncRNAs), are key components. The first three have been extensively investigated. Even though represent vast majority cellular RNA species, class remains historically understudied. This partly because a lag in technological bioinformatic innovations capable identifying accurately measuring their expression. Nevertheless, recent progress domain has enabled profusion publications novel sub-types ncRNAs studies directly addressing human health disease. Introduction Upper limb (UL) impairment is common in people with multiple sclerosis (pwMS), and functional recovery of the UL a key rehabilitation goal. Technology-based approaches, like virtual reality (VR), are increasingly promising. While most VR environments task-oriented, our clinical approach integrates neuroproprioceptive ‘facilitation inhibition’ (NFI) principles. To advance this, we developed immersive software based on NFI principles targeting function sit-to-stand ability. This study aims to evaluate effectiveness this therapy compared conventional NFI-based physical pwMS. Our uniquely applies advanced imaging techniques, along biological molecular assessments, explore adaptive processes induced by rehabilitation. Methods analysis double-arm, randomised, assessor-blinded, controlled trial runs over 2 months (1 hour, times per week). PwMS mild severe disability will receive either or real-world therapy.
