Implantable Immunostimulant Microneedle Patch for Post-Surgical Prevention of Cancer Recurrence and Distant Tumor Inhibition

Cancer

challenge
in
achieving
long-term
eradication.
Here,
we
develop
biocompatible
and
implantable
immunostimulant
microneedle
patch
designed
to
suppress
local
tumor
surgery.
The
patch,
fabricated
using
methacrylate-modified
hyaluronic
acid,
incorporates
2'3'-cGAMP,
STING
agonist,
IL-2,
cytokine
approved
for
clinical
cancer
immunotherapy
that
expands
T
cells.
enables
controlled
release
of
cGAMP
induce
dendritic
cell
maturation,
antitumor
macrophage
polarization
(M1
macrophage),
priming
activation.
Simultaneously,
localized
IL-2
activates
CD8+
cells
recruits
immune
the
microenvironment.
When
combined
with
an
anti-CTLA-4
antibody,
checkpoint
blockade,
hybrid
significantly
reduces
Treg
at
surgery
sites,
enhancing
responses
effectively
inhibiting
progression
distant
tumors
both
prophylactic
therapeutic
models.
Compared
traditional
postsurgical
chemotherapy
radiotherapy,
this
patch-mediated
demonstrates
superior
efficacy
mitigating
relapse
while
offering
