Brassica oleracea var. italica (broccoli), a member of the cabbage family, is abundant with many nutrients, including vitamins, potassium, fiber, minerals, and phytochemicals. Consequently, it has been used as functional food additive to reduce oxidative stress inflammatory responses. In current study, effects sulforaphane-rich broccoli sprout extract (BSE) on response were investigated in vitro vivo. Comparative high-performance liquid chromatography analysis sulforaphane content from different extracts revealed that 70% ethanolic BSE contained more than other extracts. qPCR enzyme immunoassay analyses markedly reduced expression proinflammatory cytokines mediators, cyclooxygenase 2, interleukin (IL)-1β, IL-6, IL-1, inducible nitric oxide synthase (iNOS), tumor necrosis factor-α (TNF-α), lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Pretreatment improved survival rate suppressed alanine aminotransferase aspartate LPS-induced endotoxemic mice, while such IL-1β, TNF-α, cyclooxygenase-2, iNOS decreased dramatically liver injury model via treatment. Additionally, F4/80 immunostaining showed hepatic macrophage infiltration after injection. conclusion, may be potential nutraceutical for preventing regulating excessive immune responses disease.

Introduction Fungus-derived secondary metabolites are fascinating with biomedical potential and chemical diversity. Mining endophytic fungi for drug candidates is an ongoing process in the field of discovery medicinal chemistry. Endophytic fungal symbionts from terrestrial plants, marine flora, fauna tend to produce interesting types importance anticancer, antiviral, anti-tuberculosis properties. Methods An organic ethyl acetate extract Penicillium verruculosum sponge-derived Spongia officinalis yielded seven different which purified through HPLC. The isolated compounds averufin (1), aspergilol-A (2), sulochrin (3), monomethyl (4), methyl emodin (5), citreorosein (6), diorcinol (7). All were characterized by high-resolution NMR spectral studies. compounds', such as antimicrobial, anti-tuberculosis, subjected bioactivity screening. Results Out tested compounds, compound (1) exhibits strong anticancer activity toward myeloid leukemia. HL60 cell lines have IC 50 concentration 1.00μm, nearly significant that standard taxol. A virtual computational molecular docking approach antigens revealed binds strongly protein target alpha, beta-tubulin (1JFF), a −10.98 binding score. Consecutive OSIRIS Lipinski ADME pharmacokinetic validation has good properties score, solubility, mutagenic nature. Furthermore, (2) first report on strain. Discussion We concluded can be taken further preliminary clinical trials like animal model in-vivo studies pharmacodynamic future prospect screening validated present experimental findings.

Inflammation is an immune system response that identifies and eliminates foreign material. However, excessive persistent inflammation could disrupt the healing process. Plant-derived exosome-like nanoparticles (PDENs) are a promising candidate for therapeutic application because they safe, biodegradable biocompatible. In this study, papaya PDENs were isolated by PEG6000-based method characterized dynamic light scattering (DLS), transmission Electron Microscopy (TEM), bicinchoninic acid (BCA) assay method, GC-MS analysis, total phenolic content (TPC) 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. For in vitro test, we conducted internalization toxicity assessment, determination of nitrite concentration, assessed expression inflammatory cytokine genes using qRT-PCR RAW 264.7 cells. vivo was induced caudal fin amputation followed analysis macrophage neutrophil migration zebrafish (Danio rerio) larvae. The result showed can be well optimized differential centrifugation with addition 30 ppm pectolyase, 15% PEG, 0.2 M NaCl, which exhibited cup-shaped spherical morphological structure average diameter 168.8±9.62 nm. storage stable aquabidest 25 mM trehalose solution at -20˚C until fourth week. Chitooligosaccharide (COS), found in both insects and marine sources, has several bioactivities, such as anti-inflammation antioxidant activities. However, the mechanism of shrimp shell COS on retardation inflammatory effects is limited. Therefore, aim this study to examine aforementioned activities LPS-activated RAW264.7 macrophage cells. significantly improved cell viability at level 500 µg/mL could reduce TNF-α, NO IL-6 generations cells (p < 0.05). Furthermore, ROS formation, NF-κB overactivation, phosphorylation Erk1/2 Akt Nrf2/HO-1 LPS-exposed These results indicate that manifests anti-inflammatory activity action via NF-κB, Erk1/2, signaling with an increasing relevance for disorders.
