Abstract Microbial consortia have been considered potential platforms for bioprocessing applications. However, the complexity in process control owing to use of multiple strains necessitates an efficient population strategy. Herein, we report circuit-guided synthetic acclimation as a strategy improve biochemical production by microbial consortium. We designed consortium comprising alginate-utilizing Vibrio sp. dhg and 3-hydroxypropionic acid (3-HP)-producing Escherichia coli direct conversion alginate 3-HP. introduced genetic circuit, named “Population guider”, E. strain, which degrades ampicillin only when 3-HP is produced. In presence selection pressure, was successfully acclimated increased 4.3-fold compared that simple co-culturing during 48-h fermentation. believe this concept useful development robust consortium-based bioprocesses.

Metabolic engineering aims to improve the production of economically valuable molecules through genetic manipulation microbial metabolism. While discipline is a little over 30 years old, advancements in metabolic have given way industrial-level molecule benefitting multiple industries such as chemical, agriculture, food, pharmaceutical, and energy industries. This review describes design, build, test, learn steps necessary for leading successful campaign. Moreover, we highlight major applications engineering, including synthesizing chemicals fuels, broadening substrate utilization, improving host robustness with focus on specific case studies. Finally, conclude discussion perspectives future challenges related engineering.

Abstract Background Classical directed evolution is a powerful approach for engineering biomolecules with improved or novel functions. However, it traditionally relies on labour- and time-intensive iterative cycles, due in part to the need multiple molecular biology steps, including DNA transformation, limited screening throughput. Results In this study, we present an ultrahigh throughput vivo continuous system thermosensitive inducible tunability, which based error-prone polymerase expression modulated by engineered thermal-responsive repressor cI857, genomic MutS mutant temperature-sensitive defect fixation of mutations Escherichia coli . We demonstrated success platform β-lactamase as model, approximately 600-fold increase targeted mutation rate. Furthermore, was combined ultrahigh-throughput methods employed evolve α-amylase resveratrol biosynthetic pathway. After rounds enrichment, 48.3% improvement activity identified via microfluidic droplet screening. addition, when coupled biosensor pathway, variant 1.7-fold higher production selected fluorescence-activated cell sorting. Yeast sex-hormone whole-cell biosensors are analytical tools characterized by long-time storage and low production cost. We engineered compact β-estradiol in S. cerevisiae cells leveraging short (20-nt long) operators bound the fusion protein LexA-ER-VP64—where ER is human estrogen receptor VP64 a strong viral activation domain. Our best showed high accuracy since their recovery concentration ranged between 97.13% 104.69%. As novelty, we built on top of them testosterone that exploit conversion into aromatase enzyme—expressed together with its co-factor CPR. used our yeast strains to evaluate activity through fluorescence measurements without need for purification. Besides, set up an aromatase-inhibitors evaluation assay measure IC50 (half-maximal inhibitory concentration) candidate compounds developed screening enzymes metabolize demands only fluorescence. These two assays allow large number chemicals proteins fast economic fashion. think work will facilitate considerably throughput discovery new drugs unknown metabolic processes.

Gallic acid is a prevalent secondary plant metabolite distinguished as one of the most effective free-radical scavengers among phenolic acids. This compound also known for its cytotoxic, anti-inflammatory, and antimicrobial activities. Bulk quantities gallic are conventionally produced by hydrolysis tannins, costly environmentally hazardous process. With aim to develop more sustainable approaches, microbial bioproduction strategies have been attempted recently. To advance synthetic biology metabolic engineering microorganisms production, we characterize here transcription factor-based inducible system PpGalR/PPP_RS13150 that responds extracellular in dose-dependent manner Pseudomonas putida KT2440. Surprisingly, this does not mediate induction when used non-native host background. We show activation requires gallate dioxygenase activity encoded galA gene. The 4-oxalomesaconic acid, an intermediate acid-metabolism, identified effector molecule interacts with factor GalR mediating gene expression. Introduction along galR enables development biosensors suitable detection monitoring extracellularly using hosts such E. coli C. necator. Moreover, P. putida-based biosensor's applicability demonstrated detecting measuring extracts Camellia sinensis leaves. study reports strategy, which can be applied developing bacterial species outside genus.
