Real-time monitoring of hydroxyl radical (⋅OH) generation is crucial for both the efficacy and safety chemodynamic therapy (CDT). Although ⋅OH probe-integrated CDT agents can track production by themselves, they often require complicated synthetic procedures suffer from self-consumption ⋅OH. Here, we report facile fabrication a self-monitored agent (denoted as Fc-CD-AuNCs) incorporating ferrocene (Fc) into β-cyclodextrin (CD)-functionalized gold nanoclusters (AuNCs) via host-guest molecular recognition. The water-soluble CD served not only capping to protect AuNCs but also macrocyclic host encapsulate solubilize hydrophobic Fc guest with high Fenton reactivity in vivo applications. The highly oxygen-dependent nature of photodynamic therapy (PDT) limits its therapeutic efficacy against hypoxic solid tumors in clinics, which is an urgent problem to be solved. Herein, we develop oxygen-independent supramolecular agent that produces hydroxyl radical (•OH) by oxidizing water the presence intracellularly abundant pyruvic acid under oxygen-free conditions. A fluorene-substituted BODIPY was designed as electron donor and coassembled with perylene diimide acceptor form quadruple hydrogen-bonded agent. Detailed mechanism studies reveal intermolecular transfer charge separation upon light irradiation result efficient generation ion pairs. Under conditions, cationic radicals directly oxidize generate cytotoxic •OH, anionic electrons acid, realizing catalytic cycle. Thus, this exhibited superb photocytotoxicity even severe environments excellent vivo antitumor on HeLa-bearing mouse models. This work provides a strategy for constructing agents, opens up avenue effective PDT tumors.

Nanomedicines for drug delivery and imaging-guided cancer therapy is a rapidly growing research area. The unique properties of nanomedicines have massive potential in solving longstanding challenges existing drugs, such as poor localization at the tumor site, high doses toxicity, recurrence, immune response. However, inadequate biocompatibility restricts their clinical translation. Therefore, advanced nanomaterials with enhanced therapeutic efficiency are highly desired to fast-track translation nanomedicines. Intrinsic nanoscale covalent organic frameworks (nCOFs), suitable size, modular pore geometry porosity, straightforward post-synthetic modification via simple transformations, make them incredibly attractive future ability COFs disintegrate slightly acidic microenvironment also gives competitive advantage targeted delivery. This review summarizes recently published applications delivery, photo-immuno therapy, sonodynamic photothermal chemotherapy, pyroptosis, combination therapy. Abstract Growing concern about photothermal tumor therapy (PTT) as a promising alternative to conventional liver cancer treatment, which is treatment strategy that utilizes near‐infrared (NIR) light‐induced agents (PTAs) yield effects localize thermal damage for tumors. Herein, given the gap between experimental research and clinical application, this review seeks timely summarize highlight recent progress of PTAs used in vivo vitro last five‐year. Abstract Photoacoustic imaging (PAI)‐guided photothermal therapy (PTT) in the second near‐infrared (NIR‐II, 1000–1700 nm) window has been attracting attention as a promising cancer theranostic platform. Here, it is reported that π‐extended porphyrins fused with one or two nanographene units (NGP‐1 and NGP‐2) can serve new class of NIR‐responsive organic agents, displaying absorption extending to ≈1000 ≈1400 nm NIR‐I NIR‐II windows, respectively. NGP‐1 NGP‐2 are dispersed water through encapsulation into self‐assembled nanoparticles (NPs), achieving high conversion efficiency 60% 69%, respectively, under 808 1064 laser irradiation. Moreover, NIR‐II‐active NGP‐2‐NPs demonstrated photoacoustic responses, along photostability biocompatibility, enabling PAI efficient PTT vivo.

Abstract High‐performance theranostic systems are of paramount importance for achieving precise image‐guided cancer immunotherapy. Here, a novel nanoplatform is presented that integrates aggregation‐induced emission luminogen (AIEgen) with prussian blue (PB) nanocatalyzer robust The AIEgen dimethylamine substitution demonstrates compelling near‐infrared (NIR) light‐induced photothermal conversion and photodynamic therapy (PDT) capabilities. By incorporating into porous PBNPs, further enveloped within M1 macrophage membrane, tumor‐specific nanoagent constructed. This strategic integration effectively constrains the molecular motion AIEgen, leading to amplified NIR‐II fluorescence brightness PDT attributes. Moreover, PBNPs can catalyze tumor‐overexpressed H 2 O generate oxygen boost efficacy, PB's NIR absorption also intensifies photoacoustic imaging effect. provides comprehensive information photoimmunotherapy in orthotopic breast cancer‐bearing mice. Leveraging its potent immunogenic cell death effect, not only significantly inhibits growth, but generates whole‐cell therapeutic vaccine protect mice from tumor rechallenge.
