The inflammasome is a macromolecular protein complex that mediates proteolytic cleavage of pro-IL-1β and -IL-18 induces cell death in the form pyroptosis. Cancer is a multifactorial pathology and it represents the second leading cause of death worldwide. In recent years, numerous studies highlighted dual role gut microbiota in preserving host’s health. Gut resident bacteria are able to produce number metabolites bioproducts necessary protect gut’s homeostasis. Conversely, several subpopulations may expand during pathological dysbiosis therefore high levels toxins capable, turn, trigger both inflammation tumorigenesis. Importantly, can interact with host either modulating directly epithelium or immune system. Numerous populating bacteria, called probiotics, have been identified as protective against genesis tumors. Given their capability homeostasis, probiotics currently tested help fight cancer patients subjected chemotherapy radiotherapy. Most recently, three independent show that specific species potentiate positive outcome anti-cancer immunotherapy. The highly significant studies, uncovering tight association between tumorigenesis, well therapy, here described. Lactobacillus rhamnosus GG (LGG), most studied probiotic model cancer, also reported. Overall, according findings summarized, novel strategies integrating such LGG, conventional therapies strongly encouraged.

Inflammation in the brain accompanies several high-impact neurological diseases including multiple sclerosis (MS), stroke, and Alzheimer's disease. Neuroinflammation is sterile, as damage-associated molecular patterns rather than microbial pathogens elicit response. The inflammasome, which leads to caspase-1 activation, implicated neuroinflammation. In this study, we reveal that lysophosphatidylcholine (LPC), a molecule associated with neurodegeneration demyelination, elicits NLRP3 NLRC4 inflammasome activation microglia astrocytes, are central players LPC-activated also requires ASC (apoptotic speck containing protein CARD), caspase-1, cathepsin-mediated degradation, calcium mobilization, potassium efflux but not caspase-11. To study physiological relevance, Nlrc4-/- Nlrp3-/- mice studied cuprizone model of neuroinflammation demyelination. Mice lacking both genes show most pronounced reduction astrogliosis microglial accumulation accompanied by decreased expression LPC receptor G2A, whereas MS patient samples increased G2A. These results NLRP3, normally form distinct inflammasomes, activate an LPC-induced important microgliosis.

Abstract Inflammation affects all stages of tumorigenesis. A key signaling pathway leading to acute and chronic inflammation is through activation the caspase-1 inflammasome. Inflammasome complexes are assembled on certain nucleotide-binding domain, leucine-rich repeat–containing proteins (NLR), AIM2-like receptors, or pyrin. Of these, NLRP1, NLRP3, NLRC4, NLRP6, AIM2 influence pathogenesis cancer by modulating innate adaptive immune responses, cell death, proliferation, and/or gut microbiota. Activation inflammasome IL18 pathways largely protective in colitis-associated colorectal cancer, whereas excessive driven IL1 promotes breast fibrosarcoma, gastric carcinoma, lung metastasis a context-dependent manner. The clinical relevance inflammasomes multiple forms highlights their therapeutic promise as molecular targets.
