The intestinal epithelium serves as an essential interface between the host and microbiota, regulating innate adaptive immunity, absorption of nutrients systemic metabolism, mediating bidirectional communication with nervous system. suffers constant challenges to proteostasis machinery due its exposure dynamically changing microbial laden lumenal gut environment high secretory demand placed on multiple epithelial cell types accommodate physiology-especially goblet, enteroendocrine Paneth cells. In all cases, cells require active unfolded protein response (UPR) sustain their physiological function, main pathway that monitors adjusts function changes in environment. A specialised endoplasmic reticulum (ER) stress sensor uniquely expressed lining mucosal surfaces, termed inositol-requiring transmembrane kinase/endoribonuclease β, has specific roles homeostasis, mucus production microbiota. Chronic ER or genetic mutations affecting key UPR mediators contribute occurrence inflammatory bowel disease ulcerative colitis, addition colon cancer. Here, we review recent advances linking physiology disease. Therapeutic strategies alleviate enforce improve ageing diseases are also discussed.

Older people experience a greater incidence of lower bowel disorders, including constipation. Causes can include factors associated with growing older, such as use medications or disease, but compounded by degenerative changes within the wall. It has been suggested that latter is exacerbated loss an effective mucosal barrier to luminal contents. In human colon, little known about impact ageing on key components this barrier, namely goblet cells and mucin content.

The intestinal area is composed of diverse cell types that harmonize gut homeostasis, which influenced by both endogenous and exogenous factors. Notably, the environment intestine exposed to several mechanical forces, including shear stress generated fluid flow, compression stretch luminal contents peristaltic waves intestine, stiffness attributed extracellular matrix. These forces play critical roles in regulation proliferation, differentiation, migration. Many efforts have been made simulate actual vitro. three-dimensional organoid culture system has emerged as a powerful tool for studying mechanism epithelial barrier, mimicking rapidly renewing epithelium from stem cells (ISCs) vivo. However, many aspects how such stress, stiffness, compression, influence remain unresolved. Here, we review recent studies elucidating impact on immunity, interaction with microbiome, diseases.

Purpose: An increasing proportion of the aging population has led to a rapid increase in number elderly patients with ulcerative colitis (UC). However, molecular mechanisms by which causes UC remain unclear. In this study, we explored role aging-related genes (ARGs) pathogenesis and diagnosis prediction. Methods: Gene expression data were obtained from four independent datasets (GSE75214, GSE87466, GSE94648, GSE169568) GEO database, ARGs derived multiple public databases. After identifying UC-related ARGs, consistent clustering was performed screen subtypes, followed exploration differences immune microenvironment pathways between distinct subtypes. Next, core module screened using WGCNA then hub characterized LASSO random forest methods. Besides, associations genes, cells, key explored. Finally, levels determined dextran sulfate sodium (DSS)-induced mouse model qRT-PCR. Results: samples classified into two subtypes (1 2), displayed significant landscape JAK/STAT signaling pathways. A series machine learning algorithms used feature (ETS1 IL7R) establish diagnostic model, exhibited satisfactory efficiency. addition, these closely associated infiltration specific cells (such as neutrophils, memory B M2 macrophages) well pathway. Later, experimental validation confirmed that ETS1 markedly increased UC. Conclusion: Overall, aging, dysregulation, process are associated. The identified particularly ETS1, could serve novel biomarkers for These findings have potential enhance understanding age-related Keywords: colitis, learning, microenvironment, biomarker

Abstract In fetal patients with intestinal atresia, surgical resection often leads to short bowel syndrome, necessitating organ transplantation. Owing a shortage of donors, alternatives such as 3D‐bioprinted artificial intestines are receiving increased interest. However, the fabrication transplantable integrating tri‐cultures viable functional cells remains challenging. This study introduces an innovative method for fabricating tri‐cultured tubular mesh intestine (TTMI) myofibroblasts, endothelial, and epithelial cells. Low‐concentration gelatin methacryloyl (GelMA) bioink is employed improve cell viability, dual cooling module incorporated cool both GelMA printing area printability. To mechanical properties TTMI transplant stability, multi‐head four‐axis bioprinter used apply bioinks polycaprolactone (PCL). The final four‐layered comprises three PCL; two middle layers printed enable cell‐to‐cell interactions. technology can be fabricate well other organs consisting different cells, ultimately enhancing availability tissues transplantation therapy.

1,3-Dioleoyl-2-palmitoyl-glycerol (OPO) is a specific triglyceride in human breast milk, and it has been added to infant formula mimic milk fat. Existing studies only focused on its effects fatty acid calcium absorption, as well the intestinal microbial composition; however, of OPO early-life development intestine were still unclear. Our study explored epithelial structure barrier construction neonatal mice involvement microorganisms. supplementation significantly increased number stem cells, which turn promoted villus crypt, goblet cell Paneth differentiation. also promotes integrity by increasing expression mucin 2, lysozyme 1, tight junction proteins. Furthermore, benefits associated with higher abundance beneficial bacteria (unclassified_f_Muribaculaceae, Akkermansia, Bifidobacterium, Blautia) elevated butyrate levels. This demonstrates efficacy health beyond defecation, expands understanding biological functions OPO, application products targeting special populations, such elderly or individuals fragility injury.
