The normal bone regeneration process is a complex and coordinated series of events involving different cell types molecules. However, this impaired in critical-size/large defects, with non-unions or delayed unions remaining major clinical problem. Novel strategies are needed to aid the current therapeutic approaches. Mesenchymal stem/stromal cells (MSCs) able promote regeneration. Their beneficial effects can be improved by modulating expression levels specific genes purpose stimulating MSC proliferation, osteogenic differentiation their immunomodulatory capacity. In context, genetic engineering MSCs expected further enhance pro-regenerative properties accelerate healing. Herein, we review most promising molecular candidates (protein-coding non-coding transcripts) discuss methodologies engineer deliver MSCs, mainly focusing on vivo animal studies. Considering potential secretome for repair, topic has also been addressed. Furthermore, results studies using discussed. Finally, debate advantages limitations genetically-engineered as promoters fracture regeneration/repair.

In this study, we investigated the anti‐osteoporotic effect of two fermented milk products (FMPs) by Lactobacillus plantarum A41 and fermentum SRK414 on a rat model ovariectomy‐induced post‐menopausal primary osteoporosis. The FMPs increased bone volume mineral density (BMD) in ovariectomized (OVX) rats, normalized biomarkers serum. Additionally, they altered gene expression levels bone‐metabolism‐related markers. Furthermore, downregulated bone‐apoptosis‐related genes stimulated ovariectomy. Interestingly, decreased inflammation markers serum, bone, ileum colon rats. Gut bacterial populations were also affected upon FMP treatment due to increase abundance genus Faecalibacterium prausnitzii. Milk L. can exhibit effects osteoporosis via regulating used study be an ideal method that has its potential treating instead drug treatments.

Emerging knowledge indicates that non-coding RNAs, including microRNAs (miRNAs) and long-noncoding RNAs (lncRNAs), have a pivotal role in bone development the pathogenesis of bone-related disorders. Most recently, miRNAs started to be regarded as potential biomarkers or targets for various sets diseases, while lncRNAs gained attention new layer gene expression control acting through versatile interactions, also with miRNAs. The rapid RNA sequencing techniques based on next-generation (NGS) gives us better insight into molecular pathways regulated by miRNA-lncRNA network. In this review, we summarize current related function regulators genes are crucial proper metabolism homeostasis. We characterized important their signatures, relationship bone. Analysis biological lncRNAs, well network, will pave way understanding think may lead innovative diagnostic tools therapeutic approaches
