Acupuncture is an integral component of complementary and alternative medicine that has been reported to enhance sperm motility, improve semen quality, consequently augment male fertility. However, the precise mechanisms action underlying molecular pathways remain unclear. In present study, we aimed elucidate potential through which acupuncture improves reproductive function in a mouse model cyclophosphamide-induced asthenozoospermia. We collected from epididymis for analysis, serum determine gonadotropin oxidative stress marker levels, conducted histological examination testicular tissue using hematoxylin eosin (HE) terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining, observed mitochondrial morphology transmission electron microscopy (TEM). also assessed levels total iron content validated proteomic transcriptomic analysis results real-time reverse transcription-quantitative polymerase chain reaction (RT-qPCR), protein imprinting immunohistochemistry (IHC). Our indicate enhances quality asthenozoospermic mice; increases testosterone (T), follicle-stimulating hormone (FSH), luteinizing (LH) levels; attenuates damage, accumulation, injury tissues. Through analyses, identified 21 key genes, cytochrome

Transferrin Receptor 2 (TfR2) is a homolog of 1 (TfR1), involved in regulating intra and extracellular iron levels. Altered pathways have been associated with cancer onset progression; however, their role canine tumors remains poorly explored. This study investigated TfR2 immunohistochemical expression non-neoplastic testis for the first time most common types testicular tumors: intratubular seminomas (ITSEMs), diffuse (DSEMs), Leydig cell (LCTs), Sertoli (SCTs). Immunohistochemical analysis revealed differential pattern according to tumor type, high observed ITSEMs DSEMs, occasional LCTs, absence SCTs. These results suggest that may play relevant seminoma development. Furthermore, specific highlights its potential as therapeutic target, where regulation possible compensatory mechanisms warrant further investigation.

An increasing body of evidence indicates that exposure to widespread, environmental and food contaminants such as mycotoxins may cause endocrine disorders infertility. Deoxynivalenol (DON), which is a toxic secondary metabolite produced by Fusarium fungi, can lead multiple harmful effects in humans animals, hepatotoxicity, nephrotoxicity, immunotoxicity, gastrointestinal toxicity, neurotoxicity, genetic toxicity carcinogenicity. Recently, there has been growing concern about DON-induced male Exposure DON its metabolites damage the structure function reproductive organs, resulting impairment gametogenesis thus impaired fertility. Potential molecular mechanisms involve oxidative stress, inflammatory response, mitochondrial dysfunction, apoptosis, cell cycle arrest, pyroptosis, ferroptosis. Moreover, several signaling pathways, including nuclear factor-kappa B, mitogen-activated protein kinase, NLR family pyrin domain containing 3, factor erythroid 2-related 2, AMP-activated apoptotic microRNAs are involved these detrimental biological processes. Research shown antioxidants, small-molecule inhibitors, or proteins (such lactoferrin) supplementation potentially offer protective targeting pathways. This review comprehensively summarizes on mammals, underlying emphasizes potential small molecules therapeutics. In further, systematic risk assessment when at doses human health, in-depth precise mechanism investigation using emerging technologies, development more effective intervention strategies warrant urgent investigation.

Iron is a key element in spermatogenesis; its metabolic pathway the testis strictly regulated. Alterations iron metabolism are linked to various diseases, including cancer, and changes metabolism-related proteins have been observed multiple human, mouse canine tumors. There limited knowledge about non-neoplastic neoplastic testes. This study aimed explore immunohistochemical expression of molecules involved uptake storage [Transferrin Receptor 1 (TfR1), ferritin (FTH1), nuclear receptor coactivator 4 (NCOA4)] PCNA testicular samples. Non-neoplastic testes showed moderate TfR1 developing germ cells Sertoli cells, high NCOA4 cytoplasmic immunostaining occasional immunopositivity for FTH1 spermatogonia cells. In contrast, Leydig cell tumors (LCTs) Diffuse Type Seminoma (DSEM) exhibited increased TfR1, along with higher expression, suggesting need proliferation. Intratubular (ITSEM) indicating greater storage, while LCTs DSEM suggested ferritinophagy release (SCTs) only expression. These preliminary findings highlight potential molecular targets new anti-neoplastic treatments

Abstract Background Diabetes mellitus (DM)-induced testicular damage is associated with sexual dysfunction and male infertility in DM patients. However, the pathogenesis of DM-induced remains largely undefined. Methods A streptozotocin (STZ)-induced diabetic model high glucose (HG)-treated vitro were established. The histological changes testes assessed by H&E staining. Serum testosterone, iron, MDA GSH levels detected using commercial kits. Cell viability lipid peroxidation was monitored MTT assay BODIPY 581/591 C11 staining, respectively. qRT-PCR, immunohistochemistry (IHC) or Western blotting employed to detect BRD7, Clusterin, EZH2 AMPK signaling molecules. associations among DNMT3a co-IP, transcriptional regulation Clusterin methylation-specific PCR (MSP) ChIP assay. Results Ferroptosis STZ mice HG-treated GC-1spg cells, this accompanied upregulation BRD7. Knockdown BRD7 suppressed HG-induced ferroptosis, as well promoter methylation HG-inactivated cells. Mechanistical studies revealed that directly bound regulated via recruiting DNMT3a. inactivation reverses silencing-suppressed ferroptosis In vivo findings showed lack protected against diabetes-induced increasing expression activating signaling. Conclusion modulating hypermethylation an dependent manner, thereby suppressing facilitate induce diabetes-associated damage.

Ferroptosis is a type of iron-dependent regulated cell death characterized by increased bioavailability redox-active iron, loss GPX4 antioxidant capacity, and oxidation polyunsaturated fatty acid-containing phospholipids mediated reactive oxygen species (ROS). The aim this study was to evaluate the effect oxidative stress induced arachidonic acid (AA) on ferroptotic in human spermatozoa.
