Abstract The centrosome is a membrane-less organelle consisting of pair barrel-shaped centrioles and pericentriolar material functions as the major microtubule-organizing center signaling hub in animal cells. past decades have witnessed functional complexity importance centrosomes various cellular processes such cell shaping, division, migration. Unexplained infertility affects about one-third of infertile couples and is defined as the failure to identify cause despite extensive evaluation male female partners. ABSTRACT Proper connection between the sperm head and tail is critical for motility fertilization. Head–tail linkage mediated by head-tail coupling apparatus (HTCA), which secures axoneme (tail) to nucleus (head). However, molecular architecture of HTCA poorly understood. Here, we use Drosophila investigate formation remodeling throughout spermiogenesis visualizing key components this complex. Using structured illumination microscopy, demonstrate that proteins Spag4 Yuri form a ‘centriole cap’ surrounds centriole (or basal body) as it invaginates into surface nucleus. As development progresses, laterally displaced side while expands under nucleus, forming what term ‘nuclear shelf’. We next show proximal centriole-like (PCL) structure positioned nuclear shelf, functioning crucial stabilizer centriole–nucleus attachment. Together, our data indicate complex, multi-point attachment site simultaneously engages PCL, ensure proper head–tail during late-stage spermiogenesis.

Infertility affects 10–15% of couples at the age conception. Mounting evidence supports that not only are paternal factors crucial during fertilization, but also for embryogenesis. This review aims to provide some clues about contribution male reproductive success and live birth, as such contributions can be important female. Spermatozoa centriolar defects can result in abnormal zygote functions. Recently, a method to quantify spermatozoa was developed named Fluorescence-Based Ratiometric Analysis of Sperm Centrioles (FRAC). However, whether identified by FRAC functions not tested. Here, we quantified using FRAC, and centriole function assessed imaging Nucleolus Precursor Body (NPB) polarization which based on the pattern NPB polarization. Data analyzed at couple embryo levels. Subjects were divided into two groups: seven couples 62 embryos with normal centrioles versus eight 78 (140 from 15 total). Patterned statistically significant both couple- embryo-level analyses (p < 0.0001 p = 0.0024). These results suggest that may correlate centrosome via scoring. This study provides foundation for more extensive studies test FRAC's utility assessing quality.
