Abstract Fungal fermentation of food and agricultural by-products holds promise for improving sustainability security. Acrolein (ACR) present in vivo and vitro can damage proteins DNA, linking it to various chronic diseases. In this paper, ergothioneine (EGT), abundant edible mushrooms, has been studied for its ability trap ACR reaction pathway with at high temperatures using ultraperformance liquid chromatography–mass spectrometry (UPLC–MS/MS). We synthesized the adducts (EGT-ACR-1 EGT-ACR-2), elucidating their structure site through HRMS nuclear magnetic resonance. Subsequently, we proved synergistic trapping effect on of EGT when combined γ-aminobutyric (GABA) disodium 5′-guanylate (GMP) binary or ternary employing Chou–Talalay methods. Quantitative analysis revealed that GABA GMP enhanced EGT's form additional adducts. World agriculture endures an immense challenge in feeding the world's growing population face of several productivity and environmental threats. Abstract Fungal natural products, including polyketides, are a rich source of bioactive molecules. Their biosynthetic enzymes encoded within gene clusters, which often activated by specific environmental conditions. As result, many products not produced under standard laboratory Heterologous expression bypasses native regulation, enabling systematic approach for polyketide discovery. The most widely used fungal hosts product production Saccharomycetales yeasts, and filamentous Eurotiomycetes. Yeasts highly tractable but have narrow scope due to their limited secondary metabolism, while Eurotiomycetes richer metabolism more difficult engineer. In this work, we established two yeasts the genera Exophiala and Knufia as novel heterologous broad range polyketides. These combine genetic tractability yeast with metabolic robustness fungi. We developed engineering tools precise integration genome editing, allowing us heterologously express five synthases different domain architectures, one involved in biosynthesis previously undescribed 2-pyridone. One Health states that human health is indispensably connected with animal and environmental health. Traditionally, humans consume meat to get animal-sourced protein in order fulfill the goal of maintaining physical However, productions seem already not supportive current food system when facing enormous population, which potentially enhances development alternative industry. In 2023, about 28.9% world populations were exposed moderate or severe insecurity. Meanwhile, intensive livestock farming has caused serious issues on atmospheric pollution, land occupation, water consumption. To alleviate such stresses, proteins come spotlight. Currently, plant-based protein, cell-cultured mycelium-based (MBP), algal-based are main categories proteins, among MBP acquiring more popularity as a nutritional, eco-friendly, sustainable source protein.
