Circadian master clocks in the brain consist of multiple neurons that are organized into populations with different morphology, physiology, and neuromessenger content presumably functions. In most animals, these distributed bilaterally, located close proximity to visual system, synchronized by eyes light-dark cycles environment. mammals cockroaches, each two consists a core region receives information from shell which output projections originate, whereas flies several other insects, lateral dorsal regions. all cases, morning evening clock seem exist, communication between them neurons, as well connection across hemispheres, is prerequisite for normal rhythmic function. Phenomena such rhythm splitting, internal desynchronization caused "decoupling" hemispheres or decoupling certain within one hemisphere. Since contain relatively few characterized at individual level, fly particularly suited study neurons. Here, we review organization bilateral brain, focus on synaptic paracrine connections comparison insects mammals.

Animals need to integrate information across neuronal networks that direct reproductive behaviors and circadian rhythms. The Drosophila master regulatory transcription factors courtship rhythms are co-expressed. We find sex differences in the number of these fruitless (fru) Clock (Clk)-expressing neurons (fru ∩ Clk neurons) regulated by male-specific Fru. assign fru electron microscopy connectome subtypes clock neurons. discover fru-expressing post-synaptic targets Clk-expressing When activated or silenced, we observe a shortening period length. Activation also changes rate behavior is performed. examine male over 24 h activities peak at lights-on. These results reveal how subserve two processes can impact behavioral outcomes sex-specific manner.

Abstract Hormones mediate inter-organ signaling which is crucial in orchestrating diverse behaviors and physiological processes including sleep activity, feeding, growth, metabolism reproduction. The pars intercerebralis lateralis insects represent major hubs contain neurosecretory cells (NSC) that produce various hormones. To obtain insight into how hormonal regulated, we have characterized the synaptic connectome of NSC adult Drosophila brain. Identification neurons providing inputs to multiple subtypes implicates diuretic hormone 44-expressing as a coordinator physiology behavior. Surprisingly, despite most having dendrites subesophageal zone (primary taste processing center), gustatory are largely indirect. We also deciphered pathways via olfactory relayed NSC. Further, our analyses revealed substantial from descending NSC, suggesting regulate both endocrine motor output synchronize changes with appropriate behaviors. In contrast inputs, sparse mostly mediated by corazonin Therefore, additionally determine putative paracrine interconnectivity between peripheral tissues analyzing single-cell transcriptomic datasets. Our comprehensive characterization network provides platform understand complex networks they orchestrate animal physiology.

Larvae of the flesh fly, Sarcophaga similis exhibit photoperiodic responses to control pupal diapause. Although external coincidence model is applicable S. photoperiodism, it remains unknown how circadian clock system integrates day-length information. To explore mechanisms, we examined neural circuitry involving lateral neurons (LNs) and prothoracicotropic hormone (PTTH) neurons. We also effects on LN-fiber patterns in third-instar larvae. Immunohistochemistry showed that protein PERIOD neuropeptide pigment-dispersing factor (PDF) were co-localized four cells per brain hemisphere, named these PDF-LNs similis. Single-cell polymerase chain reaction backfilled from ring gland two pairs pars lateralis with contralateral axons (PL-c neurons) expressed ptth. Double labeling immunohistochemistry backfills revealed PDF-immunoreactive varicose fibers projected close PL-c short f (snpf) receptor glutamate-gated chloride channel but not pdf sNPF L-glutamate PDF acutely inhibited spontaneous firing activity The number varicosities dorsal protocerebrum was significantly higher under short-day than long-day conditions a time-dependent manner. These results suggest and/or glutamate signaling PTTH form potential circuity for diapause photoperiod modifies connectivity strength between their post- or pre-neurons circuitry.
