Specialized host–microbe symbioses canonically show greater diversity than expected from simple models, both at the population level and within individual hosts. To understand how this heterogeneity arises, we utilize squash bug, Anasa tristis , its bacterial symbionts in genus Caballeronia . We modulate symbiont bottleneck size inoculum composition during colonization to demonstrate significance of ecological drift, noisy fluctuations community due demographic stochasticity. Consistent with predictions neutral theory biodiversity, found that drift alone can account for between hosts, even when 2 strains are nearly genetically identical. When acting on competing strains, maintain genetic among different hosts by stochastically determining dominant strain each host. Finally, mediates isogenic populations a single host, along consistent gradient running anterior-posterior axis symbiotic organ. Our results structure across scales does not necessarily require host-mediated selection, as it emerge result unrelated competitors. findings illuminate processes might affect transmission, coinfection, nature, which drive evolution microbe–microbe interactions host-associated microbiomes.

Streptococcus pneumoniae is an opportunistic human pathogen responsible for over a million deaths every year. Although both vaccination programs and antibiotic therapies have been effective in prevention treatment of pneumococcal infections, respectively, the sustainability these solutions uncertain. The genome highly flexible, leading to vaccine escape resistance. This flexibility predominantly facilitated by competence, state allowing cell take up integrate exogenous DNA. Thus, it essential obtain detailed overview gene expression during competence. stressed fact that administration several classes antibiotics can lead Previous studies on competence regulon were performed with microarray technology limited incomplete set known genes. Using RNA sequencing combined up-to-date annotation, we provide updated competence-regulated

Successful infection by mucosal pathogens requires overcoming the mucus barrier. To better understand this key step, we performed a survey of interactions between human respiratory and pathogen Streptococcus pneumoniae. Pneumococcal adherence to adult nasal fluid was seen only isolates expressing pilus-1. Robust binding independent pilus-1 adhesive properties but required Fab-dependent recognition RrgB, pilus shaft protein, naturally acquired secretory IgA (sIgA). Pilus-1 specific sIgA led bacterial agglutination, interaction agglutinated pneumococci entrapment in particles. test effect these vivo, were preincubated with before intranasal challenge mouse model colonization. treatment resulted rapid immune exclusion pilus-expressing pneumococci. Our findings predict that would select for nonpiliated individuals who RrgB-specific from prior episodes colonization piliated strains. Accordingly, genomic data comparing carried mothers their children showed are less likely be colonized study provides example involving antibody host, major factor driving pneumococcal adaptation.
