ABSTRACT Contrast-enhanced magnetic resonance imaging (CE-MRI) is a pivotal tool for global disease diagnosis and management. Since its clinical availability in 2009, the off-label use of ferumoxytol ferumoxytol-enhanced MRI (FE-MRI) has significantly reshaped CE-MRI practices. Unlike that enhanced by gadolinium-based contrast agents, FE-MRI offers advantages such as reduced agent dosage, extended windows, no nephrotoxicity, higher time efficiency capability molecular imaging. As leading superparamagnetic iron oxide agent, heralded next generation agents. This review delineates applications inherent technical superiority FE-MRI, providing an avant-garde medical-engineering interdisciplinary lens, thus bridging gap between demands engineering innovations. Concurrently, we spotlight emerging themes new breakthroughs. Lastly, share our own insights on potential trajectory shedding light future within medical realm.

Several FDA/EMA-approved nanomedicines have demonstrated improved pharmacokinetics and toxicity profiles compared to their conventional chemotherapeutic counterparts. The next step increase therapeutic efficacy depends on tumor accumulation, which can be highly heterogeneous. A clinical tool for patient stratification is urgently awaited. Therefore, a docetaxel-entrapping polymeric nanoparticle (89 Zr-CPC634) radiolabeled, positron emission tomography/computed tomography (PET/CT) imaging performed in seven patients with solid tumors two different doses of CPC634: an on-treatment (containing 60 mg m-2 docetaxel) diagnostic (1-2 dose (NCT03712423). Pharmacokinetic half-life 89 Zr-CPC634 mean 97.0 ± 14.4 h on-treatment, 62.4 12.9 the (p = 0.003). At these accumulation observed 46% 41% lesions median positive 96 post-injection 4.94 4.45%IA kg-1 0.91), respectively. Due to their fascinating physical, chemical, biological characteristics, nanomaterials are regarded a promising tool tackle enormous challenges in medicine. The emerging discipline nanomedicine filled huge application gap atherosclerotic field, ushering new generation diagnosis treatment strategies. Herein, based on essential pathogenic contributors atherogenesis, well distinct composition/structural synthesis strategies, surface design nanoplatforms, three major branches (nanodiagnosis, nanotherapy, nanotheranostic) elaborated. Then, state‐of‐art studies containing sequence representative significant achievements summarized detail with an emphasis intrinsic interaction/relationship between nanomedicines atherosclerosis. Particularly, attention is paid biosafety nanomedicines, which aims pave way for future clinical translation this burgeoning field. Finally, comprehensive review concluded by proposing unresolved key scientific issues sharing vision expectation future, fully elucidating closed loop from atherogenesis paradigm advancing early achievement applications.
