The endoplasmic reticulum (ER) hosts linear polypeptides and fosters natural folding of proteins through ER-residing chaperones enzymes. Failure the ER to align compose proper protein architecture leads accumulation misfolded/unfolded in lumen, which disturbs homeostasis provoke stress. Presence stress initiates cytoprotective unfolded response (UPR) restore or instigates a rather maladaptive UPR promote cell death. Although wide array cellular processes such as persistent autophagy, dysregulated mitophagy, secretion proinflammatory cytokines may contribute onset progression cardiometabolic diseases, it is well perceived that also evokes development particularly cardiovascular diseases (CVDs), diabetes mellitus, obesity, chronic kidney disease (CKD). Meanwhile, these pathological conditions further aggravate stress, creating vicious cycle. Here this review, we aimed at summarizing updating available information on CVDs, CKD, hoping offer novel insights for management comorbidities regulation

Traditional Chinese medicine plays a significant role in the treatment of various diseases and has attracted increasing attention for clinical applications. Vascular affecting vasculature heart, cerebrovascular disease, atherosclerosis, diabetic complications have compromised quality life affected individuals increase burden on health care services. Berberine, naturally occurring isoquinoline alkaloid form Rhizoma coptidis , is widely used China as folk its antibacterial anti-inflammatory properties. Promisingly, an number studies identified several cellular molecular targets berberine, indicating potential alternative therapeutic strategy vascular diseases, well providing novel evidence that supports berberine to combat diseases. The purpose this review comprehensively systematically describe agent including pharmacological effects, mechanisms, pharmacokinetics. According data published so far, shows remarkable anti-inflammatory, antioxidant, antiapoptotic, antiautophagic activity via regulation multiple signaling pathways, AMP-activated protein kinase (AMPK), nuclear factor κB (NF-κB), mitogen-activated silent information regulator 1 (SIRT-1), hypoxia-inducible 1α (HIF-1α), endothelial growth phosphoinositide 3-kinase (PI3K), B (Akt), janus 2 (JAK-2), Ca 2+ channels, endoplasmic reticulum stress. Moreover, we discuss existing limitations give corresponding measures. In addition, propose some research perspectives challenges, provide solid base from which further can excavate effective drugs monomers.

The endoplasmic reticulum (ER) is an organelle that responsible for many essential subcellular processes. Interconnected narrow tubules at the periphery and thicker sheet-like regions in perinuclear region are linked to nuclear envelope. It becoming apparent complex morphology dynamics of ER its function. Mutations proteins involved regulating structure movement implicated diseases including neurodegenerative such as Alzheimer’s, Parkinson’s, amyotrophic lateral sclerosis (ALS). also hijacked by pathogens promote their replication. Bacteria Legionella pneumophila Chlamydia trachomatis, well Zika virus, bind dynamics-regulating exploit functions advantage. This review covers our understanding morphology, functional subdomains membrane contact sites forms. We focus on current efforts quantify motion discuss related dynamics.
