The prevalence of age-related diseases has been progressively increasing worldwide. pathogenesis these disorders, including cancer, is closely associated with an age-dependent decline in cellular functions. There are multiple layers crosstalk between aging processes and oncogenesis. Therefore, a better understanding the interplay cancer will facilitate development novel therapeutic intervention strategies for malignancies at advanced ages. Recent review articles have concisely summarized updated hallmark features adequately discussed interplays both molecular levels. Because share several key mechanisms that define hallmarks, targeting shared features, such as senescence, may be beneficial prevention treatments. Notably, senolysis, innovative selectively removing senescent cells, holds great promise developing new approaches cancers other diseases, viral infections cardiovascular diseases. Herein we briefly summarize recently knowledge on well advances senolysis conditions.

Abstract Aging and cancer exhibit apparent links that we will examine in this review. The null hypothesis aging coincide because both are driven by time, irrespective of the precise causes, can be confronted with idea share common mechanistic grounds referred to as ‘hallmarks’. Indeed, several hallmarks also contribute carcinogenesis tumor progression, but some molecular cellular characteristics may reduce probability developing lethal cancer, perhaps explaining why very old age (> 90 years) is accompanied a reduced incidence neoplastic diseases. We discuss possibility process itself causes meaning time-dependent degradation supracellular functions accompanies produces byproduct or ‘age-associated disease’. Conversely, its treatment erode health drive process, has dramatically been documented for survivors diagnosed during childhood, adolescence, young adulthood. conclude connected superior including endogenous lifestyle factors, well bidirectional crosstalk, together render not only risk factor an important parameter must considered therapeutic decisions.

Abstract Caloric restriction and intermittent fasting prolong the lifespan healthspan of model organisms improve human health. The natural polyamine spermidine has been similarly linked to autophagy enhancement, geroprotection reduced incidence cardiovascular neurodegenerative diseases across species borders. Here, we asked whether cellular physiological consequences caloric depend on metabolism. We report that levels increased upon distinct regimens or in yeast, flies, mice volunteers. Genetic pharmacological blockade endogenous synthesis fasting-induced nematodes cells. Furthermore, perturbing pathway vivo abrogated lifespan- healthspan-extending effects, as well cardioprotective anti-arthritic fasting. Mechanistically, mediated these effects via induction hypusination translation regulator eIF5A. In summary, polyamine–hypusination axis emerges a phylogenetically conserved metabolic control hub for fasting-mediated enhancement longevity.

The inexorability of the aging process has sparked curiosity human beings since ancient times. However, despite this interest and extraordinary scientific advances in field, complexity hampered its comprehension. In context, Hallmarks Aging were defined 2013 with aim establishing an organized, systematic integrative view topic, which would serve as a conceptual framework for research. Ten years later promoted by progress area, updated version included three new hallmarks while maintaining original scope. review is to determine what extent achieved purpose that gave rise them. For aim, we have reviewed literature citing any two versions conclude they served not only research but also related areas knowledge. Finally, discusses candidates become part list, analyzing evidence supports whether should or be incorporated.

Cardiovascular disease (CVD) has increasing challenges for human health with an increasingly aging population worldwide, imposing a significant obstacle to the goal of healthy aging. Rapid advancements in our understanding biological process have shed new light on some important insights aging-related diseases. Although numerous reviews delved into mechanisms through which affects CVD and age-related diseases, most these relied heavily research related cellular molecular processes often observed from animal experiments. Few provided that connect hypotheses regarding patterns chronological impacts populations. The purpose this review is highlight major questions studies provide perspectives context real-world multidimensional information potential insights.

Abstract Cellular senescence and the senescence‐associated secretory phenotype (SASP) contribute to age‐related arterial dysfunction, in part, by promoting oxidative stress inflammation, which reduce bioavailability of vasodilatory molecule nitric oxide (NO). In present study, we assessed efficacy fisetin, a natural compound, as senolytic vascular cell SASP factors improve function old mice. We found that fisetin decreased cellular human endothelial culture. mice, SASP‐related inflammation were lower 1 week after final dose oral intermittent (1 on—2 weeks off—1 on dosing) supplementation. Old fisetin‐supplemented mice had higher function. Leveraging p16‐3MR transgenic model allowing genetic clearance p16 INK4A ‐positive senescent cells, ex vivo removal cells from arteries isolated vehicle‐ but not fisetin‐treated increased endothelium‐dependent dilation, demonstrating improved through senolysis. Enhanced with was mediated NO reduced cellular‐ mitochondrial‐related stress. Arterial stiffness Ex senolysis aorta rings did further mechanical wall due Lower accompanied favorable remodeling. The findings this study identify promising therapy for clinical translation target excess treat dysfunction.

Endothelial cells line at the most inner layer of blood vessels. They act to control hemostasis, arterial tone/reactivity, wound healing, tissue oxygen, and nutrient supply. With age, endothelial become senescent, characterized by reduced regeneration capacity, inflammation, abnormal secretory profile. senescence represents one earliest features ageing contributes many age-related diseases. Compared those in arteries veins, microcirculation exhibit a greater extent heterogeneity. Microcirculatory leads declined capillary density, angiogenic potentials, decreased flow, impaired barrier properties, hypoperfusion or organ-dependent manner. The heterogeneous phenotypes microvascular particular vascular bed across different tissues remain largely unknown. Accordingly, mechanisms underlying macro- micro-vascular vary pathophysiological conditions, thus offering specific target(s) for therapeutic development senolytic drugs.
