Abstract Cancers associated with pathogen infections are gradually becoming important threats to human health globally, and it is of great significance study the mechanisms carcinogenesis. Current mechanistic studies rely on animal two-dimensional (2D) cell culture models, but traditional methods have been proven insufficient for rapid modeling diseases caused by new pathogens. Therefore, research focus has shifted organoid which can replicate structural genetic characteristics target tissues or organs in vitro, providing platforms pathogen-induced oncogenic mechanisms. This review summarizes application technology four pathogen-associated cancers: gastric cancer linked Helicobacter pylori , liver hepatitis B virus C virus, colorectal Escherichia coli cervical related papillomavirus. also proposes several limitations optimize models advance treatment future.

Background Human papillomavirus (HPV) infection is a worldwide reproductive system disease. Baofukang suppository, traditional herbal preparation that includes curdione and borneol, has been reported to treat bacterial vaginosis (BV) HPV in China. However, the therapeutic mechanism still unknown. This study aims explore molecular mechanisms of borneol treating infection. Methods We conducted retrospective cohort analysis medical records from single-center involving 205 patients, focusing on correlation between clearance co-infection with other pathogens, confirming efficacy suppository. Bioinformatics network pharmacology approaches were employed identify targets suppository for BV/HPV co-infections. qRT-PCR, Western blot, immunofluorescence staining, flow cytometry utilized validate along associated immune changes. Finally, confirmed vivo using an LPS/TC-1 cervical orthotopic injection model. Results Curdione selectively inhibit secretion interleukin-6 (IL-6) interleukin-1β (IL-1β) by macrophages. The reduction IL-6 IL-1β levels effectively inhibits expression CD274 (Programmed death ligand 1, PD-L1) infected epithelial cells inhibiting STAT3 phosphorylation, thereby suppressing their evasion capabilities. Furthermore, enhance tumor necrosis factor α (TNF-α) caspase 1 (CASP1) macrophages, as well interleukin 12 (IL-12) 23 (IL-23) dendritic (DCs). these inflammatory factors promotes migration differentiation T site infection, completing cells. Conclusion main components progression occurrence cancer modulating communication innate adaptive immunity, promoting recruitment recognition CD8 + eliminate HPV-infected

Cervical cancer is the fourth leading cause of cancer-related deaths among women worldwide, causing over 660,000 new cases and 350,000 in 2022, with a disproportionately high burden low-resource countries where access to treatment limited. Human papillomavirus (HPV) common sexually transmitted infection that accounts for approximately 95% cervical cases. Persistent HPV can progress dysplasia, categorized into varying severities (CIN1, CIN2, CIN3), which significantly increases risk. The mechanism HPV-induced malignancy involves disruption cellular apoptosis by integrating viral genetic material cells, particularly within transformation zone. proteins E6 E7 play pivotal roles carcinogenesis inhibiting tumor suppressor proteins, promoting uncontrolled cell proliferation, evading immune responses, ultimately driving progression toward malignancy. Timely detection intervention are essential managing HPV-related cancers. Preventative measures such as vaccination have demonstrated substantial efficacy. Six vaccines targeting high-risk (HR) strains recommended before sexual activity or exposure. Despite these advancements, barriers, misinformation, logistical challenges, limited healthcare infrastructure, persist, underserved regions. Advances diagnostic therapeutic technologies offered avenues addressing challenges. Next-generation sequencing CRISPR gene editing emerging promising tools enable precise targeted interventions. Furthermore, artificial intelligence (AI) imaging innovations enhanced accuracy personalized care. Pap smears DNA testing indispensable early detection. To tackle globally, multifaceted approach required. Public health education, programs, research, international collaboration crucial. campaigns should combat strengthen focus on novel therapies, screening technologies, next-generation sequencing.

Human papillomavirus (HPV) is associated with a significant global burden of precancerous lesions and cancer. People HIV (PWH) are at higher risk HPV infection HPV-related diseases. This systematic review was conducted to synthesize data on the diseases among PWH. Studies published between January 2018-June 2023 were sourced from databases conferences. Included 221 publications containing epidemiological infections clinical The varied by geographical region, age, sex, sexual orientation. Compared people without (PWoH), PWH had prevalence incidence Among PWH, anal ranged 44% 83%; men anogenital warts than women. over two-fold greater transgender women who have sex their respective counterparts HIV. Incident cancer up PWoH, incident cervical six times those without. most prevalent high-risk (hr) genotypes disease vaccine genotype HPV16/18/52/58. The role of immunogenic cell death (ICD) in cervical cancer (CESC) is not well understood. This study sought to investigate the significance ICD CESC and establish an ICDRs prognostic model improve immunotherapy efficacy for patients with cancer. ICD-associated genes were screened at single-cell transcriptome levels based on AddModuleScore, single-sample gene set enrichment analysis (ssGSEA) weighted co-expression network (WGCNA) analysis. Immunogenic death-related features (ICDRs) constructed using multiple machine algorithms, evaluated training validation sets provide quantitative tools predicting prognosis clinical practice. Predictive models used risk subgroups response immunotherapy, as drug sensitivity. Finally, expression ICD-related was verified by RT-qPCR. Through integrated data, transcriptomic profiling, computational modeling, seven identified highly patients. Multivariate demonstrated that low-risk had significantly better overall survival compared high-risk patients, confirming independent tool. Assessments tumor microenvironment (TME), mutation characteristics, sensitivity within indicated a stronger group.

The present analysis provides recent trends on HR-HPV rates and distribution among 3500 Greek women between 2021 2023. Methods: detection HR-HPVs specific identification HPV16 HPV18 were conducted using Roche Cobas 4800 assay. genotyping 12 other was performed through Nested Multiplex PCR methodology (NMPCR). Results: overall rate 8.8% most prevalent genotype being followed by HPV31, HPV66, HPV56, HPV51, HPV58, HPV45, HPV18, HPV68, HPV59, HPV52, HPV35, HPV39, HPV33. Among HR-HPV-positive cases prevalence single, double, triple, quadruple infections 73.9%, 19.9%, 5.5%, 0.7%, respectively. Age-specific showed that higher in age group 31–35 years (25.5%) it estimated multiple occur more often younger women. Notably, varies different groups. It proposed HPV16, HPV66 may show an increased possibility establishing long-term over 36 old. Conclusions: high which are not included prophylactic vaccines underlines significance constant surveillance circulating HPVs population.
