Double-stranded RNA (dsRNA) binding proteins (dsRBPs) play crucial roles in various cellular processes, especially the innate immune response. Comprehensive characterization of dsRBPs is essential to understand intricate mechanisms for dsRNA sensing and Traditional methods have predominantly relied on affinity purification, favoring isolation strong binders. Here, we adopt proteome integral solubility alteration (PISA) workflow characterizing dsRBPs, resulting observation 18 known identification 200 potential dsRBPs. Next, focus zinc finger protein 385 A (ZNF385A) discover that its knockout activates transcription interferon-β absence immunogenic stimuli. The ZNF385A elevates level endogenous dsRNAs, transcripts associated with retroelements, such as short interspersed nuclear element (SINE), long (LINE), terminal repeat (LTR). Moreover, loss enhances bioactivity 5-Aza-2'-deoxycytidine (5-AZA-CdR) tumor-killing effect NK cells. Our findings greatly expand dsRBP reservoir contribute understanding homeostasis.

Pleural effusion, characterized by the accumulation of fluid in pleural space, poses significant challenges clinical practice, especially determining whether it belongs to inflammatory exudates or non-inflammatory transudates. Adenosine deaminase (ADA), an enzyme primarily produced immune cells, particularly lymphocytes, increase response conditions, including tuberculosis and malignancies. Elevated ADA levels have been shown correlate with exudates, making a valuable biomarker for differentiating between effusions. Moreover, numerous studies demonstrated treatment function inflammation- related effusion syndrome. Recently, research has established values implication diagnosing managing disease. Based on these findings, becomes reliable, non-invasive marker early diagnosis appropriate inflammation, ultimately improving patient outcomes.

The genomes of positive-sense (+) single-stranded RNA (ssRNA) viruses are believed to be subjected a wide range modifications. In this study, we focused on the chikungunya virus (CHIKV) as model ssRNA study landscape viral modification in infected human cells. Among 32 distinct modifications analysed by mass spectrometry, inosine was found enriched genomic CHIKV RNA. However, orthogonal validation Illumina RNA-seq analyses did not identify any along genome. Moreover, infection alter expression ADAR1 isoforms, enzymes that catalyse adenosine conversion. The left-handed Z-conformation of nucleic acids can be adopted by both DNA and RNA when bound Zα domains found within a variety viral innate immune response proteins. While Z-form adoption is preferred certain sequences, such as the commonly studied (CpG)n repeats, has been reported to bind wide range sequence contexts. Using NMR other biophysical techniques, find that ZαADAR1 binds LNA using similar interface for binding, dissociation constant (KD) ∼4 μM. In contrast Z-DNA/Z-RNA, two every 6 bp stretch, our data suggests multiple molecules, indicating completely different mode.
