The epithelial barrier serves as a critical defense mechanism separating the human body from external environment, fulfilling both physical and immune functions. This plays pivotal role in shielding environmental risk factors such allergens, pathogens, pollutants. However, since 19th century, escalating threats posed by pollution, global warming, heightened usage of industrial chemical products, alterations biodiversity have contributed to noteworthy surge allergic disease incidences. Notably, diseases frequently exhibit dysfunction barrier. proposed hypothesis introduces novel avenue for prevention treatment diseases. Despite increased attention development, numerous questions persist regarding mechanisms underlying disruption normal function. Consequently, this review aims provide comprehensive overview barrier’s diseases, encompassing influencing factors, assessment techniques, repair methodologies. By doing so, it seeks present innovative strategies

Abstract Background Alteration of the gut microbiota may contribute to development inflammatory bowel disease (IBD). Epigallocatechin-3-gallate (EGCG), a major bioactive constituent green tea, is known be beneficial in IBD alleviation. However, it unclear whether exerts an effect when EGCG attenuates IBD. Results We first explored oral or rectal delivery on DSS-induced murine colitis. Our results revealed that anti-inflammatory and colonic barrier integrity were enhanced by oral, but not rectal, EGCG. observed distinct EGCG-mediated alteration microbiome increasing Akkermansia abundance butyrate production. Next, we demonstrated pre-supplementation induced similar outcomes administration. Prophylactic attenuated colitis significantly enriched short-chain fatty acids (SCFAs)-producing bacteria such as SCFAs production mice. To validate these discoveries, performed fecal transplantation (FMT) sterile filtrate (SFF) inoculate DSS-treated Microbiota from EGCG-dosed mice alleviated over control SFF shown superiorly integrity, also SCFAs. Collectively, attenuation suggests intimate involvement SCFAs-producing , SCFAs, which was further prophylaxis FMT. Conclusions This study provides data indicating ameliorated inflammation microbiota-dependent manner. findings provide novel insights into remission potential modulator for prevent treat

The intestinal mucosa provides a selective permeable barrier for nutrient absorption and protection from external factors. It consists of epithelial cells, immune cells their secretions. gut microbiota participates in regulating the integrity function homeostatic balance. Genetic factors predispose individuals to dysfunction, changes composition are central this process. progressive identification these has led development concept ‘leaky syndrome’ ‘gut dysbiosis’, which underlie relationship between impairment, metabolic diseases autoimmunity. Understanding mechanisms underlying process is an intriguing subject research diagnosis treatment various extraintestinal diseases.

Autoimmune diseases are a diverse group of conditions characterized by aberrant B cell and T reactivity to normal constituents the host. These occur widely affect individuals all ages, especially women. Among these diseases, most prominent immunological manifestation is production autoantibodies, which provide valuable biomarkers for diagnosis, classification disease activity. Although cells have key role in pathogenesis, they technically more difficult assay. In general, autoimmune results from an interplay between genetic predisposition environmental factors. Genetic autoimmunity complex can involve multiple genes that regulate function immune populations. Less frequently, result single-gene mutations regulatory pathways. Infection seems be common trigger disease, although microbiota also influence pathogenesis. As shown seminal studies, patients may express autoantibodies many years before appearance clinical or laboratory signs — period called pre-clinical autoimmunity. Monitoring autoantibody expression at-risk populations therefore enable early detection initiation therapy prevent attenuate tissue damage. Autoimmunity not static, however, remission achieved some treated with current agents. host constituents. This Review provides overview basis focus on given their as markers

Environmental exposure plays a major role in the development of allergic diseases. The exposome can be classified into internal (e.g., aging, hormones, and metabolic processes), specific external chemical pollutants or lifestyle factors), general broader socioeconomic psychological contexts) domains, all which are interrelated. All factors we exposed to, from moment conception to death, part exposome. Several hundreds thousands new chemicals have been introduced modern life without our having full understanding their toxic health effects ways mitigate these effects.
