Discovery and identification of a novel yeast species, Hanseniaspora drosophilae sp. nov., from Drosophila in Okinawa, Japan DOI

Hanseniaspora , a genus of yeasts in which many species reproduce sexually, has attracted the attention researchers because its prevalence diverse ecological niches. Building on our extensive collection efforts since 2020, three previously unknown yeast strains from wild Drosophila trapped ripe bananas Okinawa, Japan, were isolated. Using multifaceted approach, including physiological assessments and sequence analysis D1/D2 domain 26S LSU rRNA gene internal transcribed spacer (ITS) region, it was revealed that these are novel members . The strains, JCM 36741 T 36742 36748, had identical sequences their respective ITS regions, justifying classification as single species. Moreover, new exhibited remarkable degree divergence closest relatives, differing by 7 nucleotide substitutions (1.28%) domain, 29 4 gaps (4.08%) regions. Agricultural crop productivity relies on the application of chemical pesticides to reduce pest and pathogen damage. However, also pose a range ecological, environmental economic penalties. Drosophila suzukii (Matsumura) or spotted wing is a worldwide invasive pest of soft- and stone-fruit production. Female D. lay their eggs in ripening fruit the hatched larvae damage from inside, rendering it unmarketable causing significant economic loss. Current methods to reduce population field primarily rely on chemical insecticides which are not sustainable long-term solution increase risk resistance developing. The invasive pest, Drosophila suzukii attacks fresh soft-skinned fruit. Broad-spectrum insecticides are implemented for control but there is a need to reduce environmental risks and insecticide residues on fruits. Hanseniaspora uvarum yeast frequently found ripe fruits associated with D. suzukii. We aim exploit the ecological association attraction of H. by developing an attract-and-kill strategy, spray-application canopy not therefore investigated attraction, egg-laying mortality when exposed insecticidal yeast-based formulations.Hanseniaspora strongly attracted applied leaves grapevine, Vitis vinifera. Abstract BACKGROUND Bactrocera cucurbitae (Coquillett) is a distructive quarantine insect pest that causes significant economic losses on cucurbit crops. To explore green control approach, we investigated the behavioral responses of B. larvae and adults to bacterial suspensions, sediments, supernatants derived from eight gut microbial strains across four distinct genera. The proboscis extension response was used evaluate impact these strains. In addition, using food selection experiments, two‐choice trap methods, gas chromatography–mass spectrometry, isolated identified predominant volatile compounds in microbiota supernatants. RESULTS Among tested strains, Kluyvera , Morganella Providencia exhibited notable attraction toward B . particular, supernatant M38 revealed most highly attractive effect larvae, whereas M72 adults. Primary components present were dimethyl disulfide, indole, 2‐nonone, phenethyl alcohol, 1‐decanol.
