Non-suicidal self-injury (NSSI) is a highly prevalent phenomenon in adolescence, often associated with prior traumatic experiences. The development and maintenance of NSSI dysregulation the stress response, evidence suggests that hypothalamic-pituitary-adrenal (HPA) axis plays an important role. endocannabinoid system neuromodulatory close functional interaction HPA axis. However, role children adolescents less clear, previously no study examined endocannabinoids youth experiences maltreatment. N-arachidonyl ethanolamide (AEA) 2-arachidonyl glycerol (2-AG) were quantified alongside sociodemographic clinical characteristics n = 148 (12–17 years age). The quantification of endocannabinoids in biological fluids is becoming increasingly popular as an indicator psychological and physiological function. Numerous methods to quantify the endocannabinoid ligands have been published so far, yet their concentrations responses often exhibit significant variability across studies. Endocannabinoids regulate interact with a wide range biomolecules, causing vary between cohorts individuals, sensitivities them depend on pre-experimental behaviours activities. Moreover, matrix effects produced by complex nature biofluids necessitate rigorous sample preparation techniques, all which introduce opportunities for both inter- intra-assay variability. This review aims address causes prior mass spectrometric analysis, including biofluid choice, human variability, collection extraction methods. If these factors are fully considered standardised introduced, may become more reliable, allowing utility clinical markers progress.

Abstract Background Major Depressive Disorder (MDD) and Alcohol Use (AUD) are two high-prevalent conditions where the Endocannabinoid system (ECS) is believed to play an important role. The ECS regulates how different neurotransmitters interact in both disorders, which crucial for controlling emotions responses stress reward stimuli. Abstract Background Posttraumatic stress disorder (PTSD) is one of the most sex-polarized psychiatric disorders, with women exhibiting twice prevalence men. The biological mechanisms underlying this sex disparity are not fully understood. Growing evidence suggests that alterations stress-buffering endocannabinoid (eCB) system and heightened inflammation central to PTSD pathophysiology may contribute sex-biases in risk severity. Here, we examined sex-differences levels circulating eCBs peripheral pro-inflammatory markers a cohort men non-psychiatric controls. Methods 88 individuals 85 sex- age-matched controls (HC) were retrospectively selected from Mass General Brigham Biobank. Serum was assayed measure [anandamide (AEA), 2-arachidonyl glycerol (2-AG), oleoylethanolamide (OEA), arachidonic acid (AA] inflammatory [interleukin-1β (IL-1β), IL-6, IL-8, IL-18, tumor necrosis factor-alpha (TNFα), C-reactive Protein (CRP)]. Linear regression used predict differential abundance each analyte by disease state (PTSD/HC) within male female subgroups. Two-sided t-tests Benjamini-Hochberg correction examine differences across Analyses repeated those comorbid major depressive disorder. Results Male patients showed significant decrease AEA, AA OEA compared ( p ’s < .001) subgroups (PTSD HCs) p< .001 ). In contrast, among females, elevated IL-6 p<.05 ) IL-8 Both exhibited an increase TNFα concentrations ), HCs. Similar results obtained subgroup MDD after controlling for FAAH 385A genotype. Conclusions Our findings show first time eCB absent PTSD, who turn exhibit broader markers, thus suggesting perturbations vary sex.

Substance use disorder (SUD) is a major global public health challenge, frequently co-occurring with psychiatric conditions such as attention-deficit/hyperactivity (ADHD). Endocannabinoid system (ECS) dysregulation has been implicated in both SUD and ADHD, but the interplay between these remains poorly understood. This study investigates plasma concentrations of endocannabinoid-congeners individuals SUD, without comorbid to identify potential biomarkers. exploratory included 469 participants divided into three groups: (1) healthy controls (n = 136), (2) patients ADHD 267), (3) 66). Plasma 12 endocannabinoid-related molecules, including acylglycerols (2-AG, 2-LG, 2-OG) acylethanolamides (AEA, DEA, DHEA, DGLEA, LEA, OEA, PEA, POEA, SEA), were quantified using high-performance liquid chromatography-mass spectrometry (HPLC-MS/MS). A multinomial Elastic Net regression model was applied assess their biomarker potential. Patients exhibited significantly lower 2-AG 2-LG compared controls, while most elevated, except for POEA SEA. comorbidity associated 2-AG, AEA, SEA, PEA elevated. Machine learning analysis identified SEA key biomarkers, achieving an accuracy 72.1 % ROC-AUC 0.77. suggests distinct ECS alterations highlighting Future research should validate findings larger cohorts explore ECS-targeted therapeutic interventions dual-diagnosis populations.

Abstract Despite the great relevance of neurosteroid allopregnanolone and related isomers to various health conditions, quantification typically involves immunoassay, which suffers from serious issues with cross-reactivity closely molecules. This article describes development partial validation a liquid chromatography coupled tandem mass spectrometry assay for simultaneous allopregnanolone, pregnanolone, isopregnanolone, epi-allopregnanolone, testosterone in human serum healthy males females aged 5–85 years. 1-amino-4-methylpiperazine (AMP) was used as derivatisation reagent enhance ionisation signal. Linearity calculated at 0.99 lower limit 10.08 pg/mL along linearity 0.98 42.32 testosterone. Application method showed sex age effects across lifespan both testosterone, whereas comparative immunoassay not able detect differences same samples. Our this should provide new tool researchers discover role its health, how it compares hormones.
