We present a brief overview of the incentive sensitization theory addiction. This posits that addiction is caused primarily by drug-induced in brain mesocorticolimbic systems attribute salience to reward-associated stimuli. If rendered hypersensitive, these cause pathological motivation (‘wanting’) for drugs. address some current questions including: what role learning and addiction? Does occur human addicts? Is development addiction-like behaviour animals associated with sensitization? What best way model symptoms using animal models? And, finally, are roles affective pleasure or withdrawal

ABSTRACT A comprehensive understanding of the neurobiology alcohol cue reactivity is critical in identifying neuropathology use disorders (AUD) and developing treatments that may attenuate craving reduce relapse risk. Functional neuroimaging studies have identified many brain areas which cues elicit activation. However, extant included relatively small numbers cases, with AUD varying severity, employed different paradigms. We used activation likelihood estimation, a quantitative, coordinate‐based meta‐analytic method, to analyze activated by alcohol‐related across studies, examine whether these were differentially between cases controls. Secondarily, we reviewed correlations behavioral measures cue‐elicited activation, as well treatment effects on such Data analyzed from 28 679 174 Among elicited robust limbic prefrontal regions, including ventral striatum, anterior cingulate ventromedial cortex. As compared controls, demonstrated greater parietal temporal posterior cingulate, precuneus superior gyrus. Cue‐elicited striatum was most frequently correlated reduced treatment, but results often derived region‐of‐interest analyses interrogated only regions. These findings support long‐standing theories mesolimbic involvement processing, suggest other more clearly differentiate Prevention for should consider interventions areas.

The rat prelimbic prefrontal cortex and nucleus accumbens core are critical for initiating cocaine seeking. In contrast, the neural circuitry responsible inhibiting seeking during extinction is unknown. present findings using inhibition of selected brain nuclei with GABA agonists show that suppression produced by previous training required activity in infralimbic cortex. Conversely, reinstatement drug a injection extinguished animals was suppressed increasing neuronal glutamate agonist AMPA. induced inactivating resembled other forms reinstated depending on basolateral amygdala. The main characteristics of cocaine addiction are compulsive drug use despite adverse consequences and high rates relapse during periods abstinence. A current popular hypothesis is that due to drug-induced neuroadaptations in reward-related learning memory processes, which cause hypersensitivity cocaine-associated cues, impulsive decision making abnormal habit-like learned behaviours insensitive consequences. Here, we review results from studies on the effect exposure selected signalling cascades, growth factors physiological processes previously implicated neuroplasticity underlying normal memory. These include extracellular signal-regulated kinase (ERK) pathway, brain-derived neurotrophic factor (BDNF), glutamate transmission, synaptic plasticity (primarily form long-term potentiation depression, LTP LTD). We also discuss degree these cocaine-induced changes mesolimbic dopamine system mediate psychomotor sensitization cocaine-seeking behaviours, as assessed animal models addiction. Finally, speculate how may interact initiate sustain seeking.
