Calix[n]arenes (n = 4, 5, 6, 8) are "chalicelike" phenol-based macrocycles that among the most fascinating and highly studied scaffolds in supramolecular chemistry. This stems from functional tunable diversity at both their upper lower rims, preorganized nonpolar cavities ion-binding sites, well-defined conformations. Conjugation of calixarene with various fluorogenic groups has led to development smart fluorescent probes have been utilized as molecular sensors, bioimaging, for drug gene delivery, self-assembly/aggregation, materials. The fine-tuning incorporation different ligating sites calix[4]arene scaffold produced numerous sensors cations, anions, biomolecules. Moreover, aqueous solubility p-sulfonatocalix[4]arenes engendered potential use drug/gene delivery enzymatic assays. In addition, because strong optical properties, calix[4]arenes used develop materials, including gels well nonlinear optical, organic light-emitting diode, multiphoton Finally, significant developments utility higher calixarenes made bioapplications. review critically summarizes recent advances all these areas.

Magnetic hyperthermia (MHT) is a therapeutic modality for the treatment of solid tumors that has now accumulated more than 30 years experience. In ongoing MHT clinical trials brain and prostate tumors, iron oxide nanoparticles are employed as intra-tumoral agents under patient-safe 100 kHz alternating magnetic field (AMF) applicator. Although currently approved by FDA imaging purposes anemia, (MNPs) designed efficient must respond to specific physical-chemical properties in terms magneto-energy conversion, heat dose production, surface chemistry aggregation state. Accordingly, past few decades, these requirements have boosted development new generation MNPs specifically aimed MHT. this review, we present an overview on their assemblies produced via different synthetic routes, focusing which MNP features allowed unprecedented heating efficiency levels be achieved highlighting nanoplatforms prevent loss intracellular environment. Moreover, review advances MNP-based embrace concept multimodal therapy, aims combine with chemotherapy, radiotherapy, immunotherapy, photodynamic or phototherapy. Next, better control temperature at tumor, focus studies optimized maintain gold-standard performance also tackling aim quantitatively assess amount tumor site regulate conditions. To conclude, future perspectives guidance how advance therapy will provided.

Tumor hypoxia has proven to be the major bottleneck of photodynamic therapy (PDT) clinical transformation. Different from traditional O2 delivery approaches, here we describe an innovative binary O2-economizer (PDOE) tactic reverse hypoxia-driven resistance by designing a superoxide radical (O2•–) generator targeting mitochondria respiration, termed SORgenTAM. This PDOE system is able block intracellular consumption and down-regulate HIF-1α expression, which successfully rescues cancer cells becoming hypoxic relieves intrinsic burden tumors in vivo, thereby sparing sufficient endogenous for PDT process. Photosensitization mechanism studies demonstrate that SORgenTAM ideal intersystem crossing rate triplet excited state lifetime generating O2•– through type-I photochemistry, generated can further trigger biocascade reduce PDT's demand O2-recycble manner. Furthermore, also serves activate AMPK metabolism signaling pathway inhibit cell repair promote death. Consequently, using this two-step O2-economical strategy, under relatively low light dose irradiation, excellent therapeutic responses toward are achieved. study offers conceptual while practical paradigm overcoming pitfalls phototherapeutics.

Poor targeting of therapeutics leading to severe adverse effects on normal tissues is considered one the obstacles in cancer therapy. To help overcome this, nanoscale drug delivery systems have provided an alternative avenue for improving therapeutic potential various agents and bioactive molecules through enhanced permeability retention (EPR) effect. Nanosystems with cancer-targeted ligands can achieve effective tumor cells utilizing cell surface-specific receptors, vasculature antigens high accuracy affinity. Additionally, stimuli-responsive nanoplatforms also been as a promising strategy against tumors, these maintain their stealth feature under conditions, but upon homing cancerous lesions or microenvironment, are responsive release cargoes. In this review, we comprehensively summarize field active number studies context emerging nanoplatform development, discuss how knowledge contribute further improvements clinical practice.
