Achieving synergism, often by combination therapy via codelivery of chemotherapeutic agents, remains the mainstay treating multidrug-resistance cases in cancer and microbial strains. With a typical core–shell architecture surface functionalization to ensure facilitated targeting tissues, nanocarriers are emerging as promising platform toward gaining such synergism. Co-encapsulation disparate theranostic agents nanocarriers─from molecules imaging or photothermal modalities─can not only address issue protecting labile drug payload from hostile biochemical environment but may also optimized release synergistic effect. However, fate co-encapsulated molecules, influenced temporospatial proximity, unpredictable marred with events deleterious impact on therapeutic efficacy, including molecular rearrangement, aggregation, denaturation. Thus, more than just an art confining multiple therapeutics into 3D nanoscale space, nanocarrier, while aiming for should strive achieving harmonious cohabitation encapsulated that, despite proximity opportunities interaction, remain innocuous each other integrity. This account will inspect current progress co-encapsulation distill out key points accomplishing synergism through reciprocity.

Freezing is commonly used to extend the shelf life of meat and products but may impact overall quality those by inducing structural changes in myofibrillar proteins (MPs) through denaturation, chemical modification, encouraging protein aggregation. This review covers effect freezing on denaturation MPs terms effects ice crystallization solute concentrations, cold oxidation. Freezing-induced begins with extracellular spaces concentrations unfrozen water fraction. At typical temperatures for (lower than −18 °C), occurs, accompanied an alteration their secondary tertiary structure. Moreover, disruption muscle cells triggers release cellular enzymes, accelerating degradation To minimize severe deterioration during frozen storage meat, there a vital need use appropriate temperature below glass transition avoid fluctuations prevent recrystallization. Such understanding MP can be applied determine optimum conditions highly retained sensory, nutritional, functional qualities.

Given their safety and efficiency in protecting protein integrity, polysorbates (PSs) have been the most widely used excipients for stabilization of therapeutics years. In recent decades, however, there numerous reports about visible or sub-visible particles PS-containing biotherapeutic products, which is a major quality concern parenteral drugs. Alternative that are safe administration, efficient different drugs against various stress conditions, effective high-concentrated liquid formulations, stable under storage conditions duration product's shelf-life, compatible with other formulation components primary packaging highly sought after. The aim this paper to review potential alternative from families, including surfactants, carbohydrate- amino acid-based excipients, synthetic amphiphilic polymers, ionic liquids enable stabilization. For each category, important characteristics such as ability stabilize proteins thermal mechanical stresses, current knowledge related profile interactions components, debated. Based on provided information detailed discussion thereof, may pave way identification development

Protein-based therapeutics have revolutionized the pharmaceutical industry and become vital components in development of future therapeutics. They offer several advantages over traditional small molecule drugs, including high affinity, potency specificity, while demonstrating low toxicity minimal adverse effects. However, manufacturing processes protein-based presents challenges related to protein folding, purification, stability immunogenicity that should be addressed. These proteins, like other biological molecules, are prone chemical physical instabilities. The drugs throughout entire manufacturing, storage delivery process is essential. occurrence structural instability resulting from misfolding, unfolding, modifications, as well aggregation, poses a significant risk efficacy these overshadowing their promising attributes. Gaining insight into alterations caused by aggregation impact on for advancement refinement Hence, this review, we discussed some features during production, formulation stabilization strategies engineering computational methods prevent aggregation.
