Lectins are capable of reading out the structural information contained in carbohydrates through specific recognition processes. Determining binding epitope sugar is fundamental to understanding this event. Nuclear magnetic resonance (NMR) a powerful tool obtain solution; however, when involved complex oligosaccharide, such as high mannose, signal overlap found NMR spectra precludes an accurate analysis interaction. The introduction tags into these oligosaccharides could overcome problems and facilitate studies. Here, we show preparation Man9 mannose with some fluorine study interaction its receptor, dendritic cell-specific intercellular adhesion molecule-3-grabbing nonintegrin (DC-SIGN). This fluorinated ligand has allowed us apply heteronuclear two-dimensional (2D) 1H,19F STD-TOCSYreF experiments, using initial slope approach, which facilitated Man9/DC-SIGN interaction, unequivocally providing epitope.

Abstract The process of protein transport across membranes involves a variety factors and has been extensively investigated. Traditionally, proteinaceous translocons chaperones have recognized as crucial in this process. However, recent studies highlighted the significant roles played by lipids glycolipid present biological membrane transport. Membrane can influence efficiency altering physicochemical properties membranes. Notably, our revealed that diacylglycerol (DAG) attenuates mobility core region, leading to dramatic suppression integration. Conversely, Escherichia coli inner membranes, named integrase (MPIase), enhances integration not only through alteration but also via direct interactions with proteins. This review explores mechanisms mediated lipids, specifically DAG, MPIase. Our results, along employed analysis methods such fluorescence measurements, nuclear magnetic resonance, surface plasmon docking simulation, are presented elucidate these mechanisms.
