Abstract Background/objectives The present study investigated the effect of curcumin and eicosapentaenoic acid, as one main components omega-3 polyunsaturated fatty acids, on anthropometric, glucose homeostasis, gene expression markers cardio-metabolic risk in patients with type 2 diabetes mellitus. Subjects/methods This clinical trial was conducted at Endocrinology Clinic Imam Reza Hospital Tabriz. Background The effects of resveratrol supplementation on inflammation and oxidative stress in patients with type 2 diabetes mellitus (T2DM) were controversial. This study explores the potential of curcumin (CUR), extracted from Curcuma longa , in combating obesity and Type 2 diabetes. Obesity diabetes were induced rats through a high‐fat high‐fructose diet (HFFD), CUR, after purification characterization by Fourier transform infrared spectroscopy (FTIR) ultraviolet (UV) spectroscopy, was administered for 3 months via gastric gavage. The results show that CUR supplementation activates insulin signaling pathway dose‐dependent manner, leading to improved sensitivity. Specifically, administering at daily dose 100 mg/kg significantly reduces activities protein tyrosine phosphatase (PTP1B) dipeptidyl peptidase‐4 (DPP‐4) 43% 45%, respectively, obese diabetic compared untreated rats. Furthermore, effectively inhibits lipase α ‐amylase both serum intestinal levels. In rats, administration glycogen phosphorylase (GP) activity 35% enhances synthase (GS) 78%, substantial increase hepatic content. Additionally, also led 21% reduction food intake 12% decrease water consumption.
