MicroRNAs (miRNAs) are small noncoding RNAs that regulate gene expression and implicated in various diseases, including cancer. Due to their critical role diagnostics, there is a growing need for sensitive, specific, rapid detection methods miRNAs. In this study, we present dual-accelerated signal amplification platform miRNA biosensing, which integrates spatial confining catalytic hairpin assembly (SC-CHA) with spherical CRISPR/Cas12a (S-CRISPR/Cas12a) system (SC-CHA@S-CRISPR/Cas12a) trans-cleavage of DNA reporters. The method employs biotinylated palindrome-rich sequence (PAS) form nanoballs, serve as scaffold the operation SC-CHA upon binding. products bind crRNA Cas 12a protein, activating S-CRISPR/Cas12a cleave reporter generate detectable fluorescence signal. uniqueness lies combined use nanoballs reporters, both significantly accelerate reaction kinetics, resulting generation.
