Abstract Background Astaxanthin (AST) exists as an ‘all‐ E ’ isomer and various ‘ Z isomers due to its multiple conjugated double bonds. The configuration has been reported exhibit stronger antioxidant activity, raising interest in potential health benefits. All‐ AST is a potent chemopreventive agent for weight loss but the anti‐obesity efficacy of remains unclear. This study therefore explored role geometric (all‐ , 9‐ 13‐ ) on alleviation obesity using Caenorhabditis elegans well established model organism. Results reduced high‐fat worms substantially. Triglyceride (TG) decreased by 18.84%, with all‐ AST, 41.18%, 35.94% ( P < 0.05), indicating that displayed superior effect. Interestingly, particularly ‐isomers, large lipid droplets (LDs) oleic acid/stearic acid (C18:1Δ9/C18:0) significantly; these contribute accumulation. also minimized food intake boosted energy consumption, ‐isomers outperformed enhancing mobility. Using quantitative polymerase chain reaction (qPCR), green fluorescent protein binding, gene‐deficient nematodes, findings current indicated especially isomers, suppressed key gene expression sbp‐1 / mdt‐15 insulin/insulin‐like growth factor signaling (IIS) pathways, which are responsible regulating fatty composition balance, subsequently co‐downregulated essential genes fat‐6 fat‐7 involved C18:1 Δ 9 synthesis. Conclusion provides important insights into association between effects carotenoids, guiding their application field. © 2025 Society Chemical Industry.

Carotenoids constitute compounds of significant biological interest due to their multiple activities, such as antimicrobial, anticancer, antiadipogenic, antidiabetic, and antioxidant properties. Metabolic syndrome (MetS) comprehends a series metabolic abnormalities (e.g., hypertension, obesity, atherogenic dyslipidemia) that can affect children, adolescents, the elderly. The treatment MetS involves numerous medications, which, despite efficacy, pose challenges prolonged use, high costs, various side effects. derivatives have been proposed alternative treatments because they reduce serum triglyceride concentrations, promote insulin response, inhibit adipogenesis, downregulate angiotensin-converting enzyme activity. However, carotenoids are notably sensitive pH, light exposure, temperature. This review addresses activity lycopene, lutein, fucoxanthin, astaxanthin, crocin, β-carotene towards MetS. It includes discussion sources, extraction methods, characterization techniques for analyzing carotenoids. Encapsulation approaches critically reviewed alternatives prevent degradation improve performance A brief overview physiopathology epidemiology diseases, including MetS, is also provided.
