Rational Design of NIR‐II Fluorescence/Photoacoustic Nanosensor Tailored for Mechanisms of Diabetes‐Related Breast Cancer

Breast
cancer
(BC)
is
the
second
most
common
cause
of
induced
death
worldwide.
Current
statistics
has
disclosed
that
diabetic
BC
patients
have
significantly
worse
survival
rate
compared
with
nondiabetic
patients.
However,
specific
mechanism
still
being
explored.
Herein,
a
novel
NIR-II
nanosensor
DNPS
for
nitric
oxide
(NO)
fluorescence/photoacoustic
(FL/PA)
imaging
capability
developed
to
explore
by
which
diabetes
promoting
breast
progression.
In
model,
exhibits
great
advantages
low
intrinsic
background,
high
sensitivity,
and
deep
tissue
penetration
successfully
confirmed
expression
level
NO
higher
than
indicating
causes
elevated
levels
in
tumor
microenvironment.
RNA-seq
analysis
results
show
hyperglycemia
caused
leads
weakened
immune
response
initiates
transcription
translation
inducible
synthase
(iNOS)
gene
produce
NO.
Besides,
increased
carcinogens
related
Nitric
2
(Nos2),
such
as
Spp1,
Mmp11,
Kitl,
develop
more
rapidly.
Here,
probe
applied
first
study
diabetes-related
certain
reference
value
provided
subsequent
research
on
progression
cancer.

Flexible Regulation of Optical Properties Based on Structure Size‐Driven Intermolecular Interactions for Phototherapy

Abstract
The
precise
control
of
optical
properties
in
molecular
systems
remains
a
challenge
for
phototherapy.
Herein,
the
strategic
combination
aggregation‐caused
quenching
(ACQ)
and
aggregation‐induced
emission
(AIE)
molecule
creates
ACQ@AIE
bimolecular
with
tunable
properties,
which
are
almost
unattainable
by
single‐component
materials.
Through
systematic
investigation
three
systems,
it
is
established
that
structure
size
differentials
dictate
their
intermolecular
interactions
consequent
behaviors.
Crucially,
AIE
smaller
promotes
ACQ
clustering
to
enhance
photothermal
effect,
while
when
becomes
larger,
particularly
approaching
molecule,
facilitating
π–π
stacking
boosting
photodynamic
effect.
These
distinct
assembly
modes
revealed
through
combined
experimental
theoretical
analyses,
enable
regulation
versus
effects
simply
regulating
ratio
molecules.
Building
on
these
mechanistic
insights,
optimal
system
engineered
into
nanoparticles
exhibit
mild
strong
excellent
tumor
accumulation
retention,
achieving
near‐complete
eradication
minimal
treatment
cycles
maintaining
good
biosafety.
This
work
not
only
elucidates
fundamental
size‐interaction‐property
relationships
but
also
provides
generalizable
strategies
developing
intelligent
photo
theranostic
materials
controlled
interaction.

Achieving
accurate
and
efficient
tumor
imaging
is
crucial

optical
technology
within
second
near-infrared
(NIR-II)
window
has
garnered
significant
interest
for
its
remarkable
benefits,
such
enhanced
tissue
penetration
depth,
superior
signal-to-background
ratio
(SBR),
minimal
autofluorescence,
reduced
photon
attenuation,
lower
scattering.
This
review
explained
design
optimization
nano-agents
responsive
to
NIR-II
window,
single-walled
carbon
nanotubes,
quantum
dots,
lanthanum-based
nanomaterials,
noble
metal
nanomaterials.
These
enable
non-invasive,
deep-tissue
with
high
spatial
resolution
their
properties
significantly
improve
accuracy,
efficiency,
versatility
imaging-guided
treatments.
And
we
discussed
characteristics
advantages
fluorescence
(FL)/photoacoustic
(PA)
providing
a
comprehensive
overview
latest
research
progress
different
FL/PA
therapy.
Furthermore,
exhaustively
reviewed
applications
multifunctional
nano-phototherapy
technologies
carried
out
by
light
including
photothermal
therapy
(PTT),
photodynamic
(PDT),
combined
modalities
like
photothermal-chemodynamic
(PTT-CDT),
photothermal-chemotherapy
(PTT-CT),
photothermal-
immunotherapy
(PTT-IO).
integrated
approaches
have
gradually
matured
over
past
decade
are
expected
become
safe
effective
non-invasive
treatment.
Finally,
outlined
prospects
challenges
development
innovation
diagnosis
nanoplatform.

future
advancements
treatment
platforms.
