The mammalian target of rapamycin (mTOR) is a central regulator diverse array cellular processes, including cell growth, proliferation, autophagy, translation, and actin polymerization. Abstract Single-cell analysis reveals aspects of cellular physiology not evident from population-based studies, particularly in the case highly multiplexed methods such as mass cytometry (CyTOF) able to correlate levels multiple signalling, differentiation and cell fate markers. Immunofluorescence (IF) microscopy adds information on morphology microenvironment that are obtained using flow-based techniques, but multiplicity conventional IF is limited. This has motivated development imaging require specialized instrumentation, exotic reagents or proprietary protocols difficult reproduce most laboratories. Here we report a public-domain method for achieving high single-cell cyclic immunofluorescence (CycIF), simple versatile procedure which four-colour staining alternates with chemical inactivation fluorophores progressively build multichannel image. Because CycIF uses standard instrumentation no more expensive than IF, it suitable high-throughput assays screening applications.
