Abstract
Pathogenic
variants
of
the
J-domain
protein
DNAJC12
cause
parkinsonism,
which
is
associated
with
a
defective
interaction
tyrosine
hydroxylase
(TH),
rate-limiting
enzyme
in
dopamine
biosynthesis.
In
this
work,
we
characterize
formation
TH:DNAJC12
complex,
showing
that
binding
stabilizes
both
TH
and
variant
TH-p.R202H,
deficiency.
This
delays
their
time-dependent
aggregation
an
Hsp70-independent
manner,
while
preserving
activity
feedback
regulatory
inhibition
by
dopamine.

barely
activates
Hsc70
but
synergistically
stimulates
ATPase
when
complexed
TH.
Cryo-electron
microscopy
supported
crosslinking-mass
spectroscopy
reveals
two
monomers
bound
per
tetramer,
