Diabetic gastroparesis (DGP) is a common complication of diabetes mellitus, marked by gastrointestinal motility disorder, delayed gastric emptying present in the absence mechanical obstruction.Clinical manifestations include postprandial fullness and epigastric discomfort, bloating, nausea, vomiting.DGP may significantly affect quality life productivity patients.Research on relationship between dynamics DGP has received much attention because increasing prevalence DGP.Gastrointestinal disorders are closely related to variety factors including destruction interstitial cells Cajal, abnormalities neuro-endocrine system hormone levels. Therefore, this study will review recent literature mechanisms as well development prokinetic treatment order give future research directions identify strategies for DGP. Slow transit constipation (STC) is a long-lasting and prevalent intestinal condition, marked by hard, dry feces. The primary cause of STC may be attributed to an imbalance in the gut's microbial community alterations its metabolic byproducts. Tongbian formula (TB), traditional Chinese medicinal formula, has been used treat shows great effect on relieving constipation. role TB regulating microbiota not fully elucidated. Herein, we investigated potential gut further explored mechanism behind effects. Our study demonstrated that significantly increased fecal water content ink propulsion rate loperamide (Lope)-induced rats. 5-HT signaling was suppressed colon tissue, abundance butyric acid (BA) colonic contents down-regulated after Lope treatment. Notably, administration led restoration dysbiosis up-regulation BA content, subsequently activating pathways. When combined with tryptophan hydroxylase-1 (TPH1) inhibitor, which crucial for synthesis, therapeutic efficacy treating compromised. alleviates reversing through increasing levels. These findings suggest ideal candidate

Chronic migraine (CM) is a highly disabling and burdensome disease. Wuzhuyu decoction (WZYD), clinical used formula to treat prevent episodic CM, has been reported relieve the hyperalgesia of CM increase brainstem blood serotonin (5-hydroxytryptamine, 5-HT) in model rats previous studies; yet mechanism unclear. This study aimed observe relief effect WZYD investigate mechanistic association with regulation on central peripheral 5-HT.WZYD different doses (3.372, 1.686 0.843 g/kg∙d) positive drug - sumatriptan (5.83 mg/kg∙3 d) were intragastrically administered inflammatory soup (IS)-induced rats, respectively. Hyperalgesia was assessed by facial mechanical withdrawal threshold tail-flick latency. 5-HT determined ELISA. Western blot analysis, immunohistochemistry immunofluorescence determination, 16S rRNA gene sequencing performed.WZYD significantly relieved elevating In groups, increased decreased calcitonin gene-related peptide both plasma, downregulated TNF-α, IL-1β, c-fos expression observed dose-dependent manner. Interestingly, colon tissues also observed, which associated upregulating tryptophan hydroxylase, transporter Piezo1 increasing chromogranin A enterochromaffin cells. Disorder microbiota, function metabolism correlated synthesis. could regulate abundance Anaerostipes Acidifaciens.WZYD pharmacological relieving possibly affecting 5-HT.
