ABSTRACT Microplastic (MP) pollution has emerged as a significant environmental and health concern. This study aimed at developing novel method for detecting MP particles in deparaffinized formalin-fixed paraffin-embedded (FFPE) routine tissue sections of human colon samples. By utilizing mid-infrared photothermal (MIP) microscopy, also known optical infrared (OPTIR) spectroscopy, we here to establish work-flow representing non-destructive spectroscopic approach distinct identification localization polymers FFPE by respecting standardized protocols well integrated into clinical routine. After OPTIR analysis, samples were processed H&E staining histopathological analysis defined regions interest. using succeeded localizing polyethylene (PE), polystyrene (PS), terephthalate (PET) MPs interest (21 PE particles, 1 PS particle, PET fiber) clearly identifying the embedded respective sections. Subsequent confirmed characteristic features inflammation spatially associated with polymers. We present first time diagnostic workflow that potential enhance our understanding accumulation organ slides allows exploration its implications health. A number of applications require methods to detect with high spatial resolution and chemical specificity microplastics (MPs) extracted from different matrices. Here we introduce a wide-field hyperspectral Fourier transform Raman microscope for the rapid detection identification MPs. The instrument, based on common-path birefringent interferometer, combines (∼1 μm) spectral (∼23 cm-1) fast measurement times (∼15 min 100 kpixel image) enables suppression sample fluorescence by proper choice scan interval interferometer. After validating instrument MPs commercial origin, demonstrate its ability matrices, filtering seawater pretreated gastrointestinal tracts fish, analyzing concentrated onto filters. Vibrational spectroscopy, including Raman scattering and infrared (IR) absorption, provides essential molecular fingerprint information, facilitating diverse applications, such as interfacial sensing, chemical analysis, biomedical diagnostics. The complementary selection rules of IR spectroscopies offer distinct, yet mutually reinforcing, insights into structure dynamics. However, in dynamic or complex environments, either technique alone is not capable providing a complete nuanced picture vibrations. Simultaneous detection vibrational modes within the same group remains challenging due to wavelength discrepancies sensitivity mismatches between spectroscopies. In this work, address gap these spectroscopies, we developed an integrated approach based on vibrational-encoded fluorescence (VEF), which information respectively encoded different parts radiation: Stokes carrying anti-Stokes reflecting information. This method employs dual-resonant microsphere-on-mirror plasmonic bridge waveband gap, enabling simultaneous visible spectrum with ultrahigh down ∼100 molecules. Hyperspectral colocalization imaging demonstrates spatial correlations By careful calibration, efficiency improved by 8 orders magnitude compared unenhanced spectroscopy. creates new opportunities for precise identification environments.
