The placenta plays a crucial role in successful mammalian reproduction. Ruminant animals possess semi-invasive characterized by highly vascularized structure formed maternal endometrial caruncles and fetal placental cotyledons, essential for full-term development. cow harbors at least two trophoblast cell populations: uninucleate (UNC) binucleate (BNC) cells. However, the limited capacity to elucidate transcriptomic dynamics of natural environment has resulted poor understanding both molecular cellular interactions between cells niches, mechanisms governing differentiation functionalization. To fill this knowledge gap, we employed Stereo-seq map spatial gene expression patterns near single-cell resolution 90 130 days gestation, attaining high-resolution, spatially resolved profiles. Based on clustering marker analyses, key transcription factors, including YBX1 NPAS2, were shown regulate heterogeneity subpopulations. Cell communication trajectory analysis provided framework cell-cell trophoblasts into BNCs microenvironment. Differential trajectories identified set genes involved regulation differentiation. Additionally, modules co-variant that help shape specific tissue structures identified. Together, these findings provide foundational insights important biological pathways critical development function cows.

Ruminants have a unique placenta in comparison to other mammalian species. Initially, they possess non-invasive epitheliochorial type of during conceptus elongation. As the trophectoderm begins attach luminal epithelium (LE) endometrium, binucleate cells (BNCs) develop within trophoblast chorion. The BNCs migrate and fuse with uterine LE form multinucleate syncytial plaques sheep hybrid trinucleate cattle. This area ruminant is semi-invasive synepitheliochorial. foundation placental cotyledons express placenta-specific genes including pregnancy-associated glycoproteins chorionic somatomammotropin hormone 2 or lactogen. Attachment interdigitation into endometrial caruncles placentomes that are subsequently vascularized provide essential nutrients for growth fetus. chapter review will discuss historical current aspects implantation development ungulates focus on cattle sheep. Supporting healthy pregnancy outcomes requires a comprehensive understanding of the molecular and cellular programs peri-implantation development, when most failure occurs. Here, we present single-cell transcriptomes bovine embryo development at day 12, 14, 16, 18 post-fertilization. We defined composition gene expression embryonic disc, hypoblast, trophoblast lineages in embryos, identified markers pathway signaling that represent distinct stages lineages; selected was validated embryos. Using detailed time-course transcriptomic analyses, revealed previously unrecognized primitive cell lineage. also characterized conserved divergence lineage between other mammalian species. Finally, established cell-cell communication underlies extraembryonic interaction to ensure proper early development. These data provide foundational information discover essential biological underpinning

Mammals differ regarding their placentae, but in all species placental trophoblasts interact intimately with the uterine endometrium to mediate transfer of nutrients from mother embryo/fetus through closely juxtaposed microcirculatory systems uterus and placenta. Placentation ruminants is intermediate between non-invasive type, as observed epitheliochorial placenta pigs, invasive haemochorial placentae mice humans. In ruminants, trophoblast cells invade endometrial tissue, invasion believed be limited luminal epithelium (LE). LE there are varying degrees syncytialisation among species, being more extensive sheep than cows. The hallmarks placentation include: (1) an extended period which conceptuses (embryos associated membranes) elongate must supported by secretions (histotroph) uterus; (2) a cascade involving array adhesion molecules that includes integrin-mediated attachment conceptus for implantation; (3) developing early placenta, process currently understanding; (4) development placentomes define cotyledonary cows sheep, provide haemotrophic support fetal development.

The uterus is vital for successful reproduction in mammals, and two different types of epithelia (luminal glandular) are essential embryo implantation pregnancy establishment. However, the cellular molecular factors pathways governing postnatal epithelium maturation, determination, differentiation developing yet to be elucidated. Here, neonatal mouse was isolated subjected single-cell transcriptome (scRNA-seq) analysis. Both undifferentiated determined luminal were heterogeneous contained several cell clusters based on transcription profiles. Substantial gene expression differences evident as matured differentiated between days 1 15. Two new glandular epithelium-expressed genes ( Gas6 Cited4 ) identified validated by situ hybridization. Trajectory analyses provided a framework understanding lineage bifurcation, differentiation. A candidate set regulatory networks that potentially direct specification morphogenesis. This atlas provides foundation important discover intrinsic mechanisms directing uterine morphogenesis during critical window development.

Syncytins are endogenous retroviral envelope proteins which induce the fusion of membranes. A human representative this group, retrovirus group W member 1 (ERVW-1) or syncytin-1 is present in trophoblast-derived extracellular vesicles and supports incorporation these into recipient cells. During pregnancy, placenta-derived participate feto-maternal communication. Bovine fetal binucleate trophoblast cells express syncytin, bovine protein K1 (BERV-K1). These release maternal stroma, but it unclear whether BERV-K1 included vesicles. Here, were isolated from placental tissue using collagenase digestion, ultracentrifugation, size exclusion chromatography.
