Disadvantaged socioeconomic position (SEP), including lower educational attainment and household income, may influence cancer risk outcomes. We hypothesized that DNA methylation could function as an intermediary epigenetic mechanism internalizes reflects the biological impact of SEP. Longstanding racial/ethnic inequalities in morbidity and mortality persist the United States. Although determinants of health are complex, social structural factors produced by inequitable racialized systems recognized as contributing sources. Social epigenetics is an emerging area research that aims to uncover biological pathways through which experiences affect outcomes. A growing body literature links adverse exposures epigenetic mechanisms, namely DNA methylation, offering a plausible pathway may arise. This review provides overview highlights existing linking exposures—i.e., psychosocial stressors, racism, discrimination, socioeconomic position, neighborhood environment—to methylation humans. As the master regulator in utero, placenta is core to Developmental Origins of Health and Disease (DOHaD) hypothesis but historically understudied. To identify placental gene-trait associations (GTAs) across life course, we perform distal mediator-enriched transcriptome-wide association studies (TWAS) for 40 traits, integrating multi-omics from Extremely Low Gestational Age Newborn Study. At [Formula: see text], detect 248 GTAs, mostly neonatal metabolic 176 genes, enriched cell growth immunological pathways. In aggregate, genetic effects mediated by expression significantly explain 4 early-life traits no later-in-life traits. 89 GTAs show significant mediation through variants, identifying hypotheses regulation GTAs. Investigation one human placenta-derived choriocarcinoma cells reveal that knockdown mediator gene EPS15 upregulates predicted targets SPATA13 FAM214A, both associated with waist-hip ratio TWAS, multiple genes involved These results suggest profound health impacts genomic developmental programming course.

Applying high-throughput DNA sequencing technologies to the ancient molecules preserved in subfossil material can provide genetic information from past individuals, populations and communities at genomic scale.
