Milk oligosaccharides are complex carbohydrates composed of various monosaccharide units linked together by glycosidic bonds. They play an essential role in promoting gut health fostering beneficial bacteria, supporting the development immune system, and protecting against infections diseases. This work compared oligosaccharide profiles widely utilized breeds such as Holstein Ayrshire (Nordic Red), with native Northern Finncattle, which is considered endangered breed. Oligosaccharides were extracted from milk analyzed liquid chromatography-mass spectrometry. In this study, we aimed to explore the protective effects of Bifidobacterium in colitis mice and potential mechanisms. Results showed that breve (B. breve) effectively colonized intestinal tract alleviated symptoms by reducing disease activity index. Moreover, B. mitigated epithelial cell damage, inhibited pro-inflammatory factors, upregulated tight junction (TJ)-proteins. Gut microbiota metabolome analysis found boosted bile acid-regulating genera (such as Clostridium sensu stricto 1), which promoted acid deconjugation intestine. Notably, cholic (CA) was closely associated with expression levels inflammatory factors TJ-proteins (p < 0.05). Our vitro experiments further confirmed CA (20.24 ± 4.53 pg/mL) contributed inhibition lipopolysaccharide-induced tumor necrosis factor-α (49.32 5.27 enhanced (Occludin Claudin-1) MUC2. Human milk oligosaccharides (HMOs) are a major component of human milk. They associated with multiple health benefits and manufactured on large scale for their addition to different food products.
