Type 2 diabetes mellitus (T2DM) patients are at a higher risk of developing Alzheimer’s disease (AD). Mounting evidence suggests the emerging important role circadian rhythms in many diseases. Circadian rhythm disruption is considered to contribute both T2DM and AD. Here, we review relationship among disruption, AD, suggest that occurrence progression AD may part be associated with disruption. Then, summarize promising therapeutic strategies targeting dysfunction for including pharmacological treatment such as melatonin, orexin, molecules, well non-pharmacological treatments like light therapy, feeding behavior, exercise.

Although circadian biorhythms of mitochondria and cells are highly conserved crucial for the well-being complex animals, there is a paucity studies on reciprocal interactions between oxidative stress, redox modifications, metabolism, thermoregulation, other major oscillatory physiological processes. To address this limitation, we hypothesize that circadian/ultradian interaction redoxome, bioenergetics, temperature signaling strongly determine differential activities sleep-wake cycling mammalians birds. Posttranslational modifications proteins by reversible cysteine oxoforms, S-glutathionylation S-nitrosylation shown to play role in regulating mitochondrial reactive oxygen species production, protein activity, respiration, metabolomics. Nuclear DNA repair cellular synthesis maximized during wake phase, whereas redoxome restored remodeling sleep. Hence, our analysis reveals wakefulness more protective restorative nucleus (nucleorestorative), sleep (mitorestorative). The "redox-bioenergetics-temperature mitochondrial-nuclear regulatory hypothesis" adds understanding respiratory uncoupling, substrate control hibernation. Similarly, hypothesis explains how redox-bioenergetics-temperature-regulated states, when perturbed interactome disturbances, influence pathogenesis aging, cancer, spaceflight health effects, sudden infant death syndrome, diseases metabolism nervous system.

Association of ARNTL and PER1 genes with Parkinson's disease: a case-control study of Han Chinese DOI Creative Commons

Abstract Circadian disruptions may result in sleep problems, oxidative stress and an altered inflammatory response. These symptoms contribute to PD pathogenesis, despite a lack of direct experimental evidence supporting this relationship. Clock genes are essential drive maintain circadian rhythm. To elucidate the possible role PD, we investigated 132 tag variants eight clock genes. We genotyped these tags within 1,394 Chinese cases 1,342 controls using Illumina GoldenGate chips. discovered that SNPs ARNTL (rs900147, P = 3.33 × 10 −5 , OR 0.80) PER1 (rs2253820, 5.30 −6 1.31) significantly associated with risk. Moreover, positive association rs900147 variant was more robust tremor dominant (TD) ( 3.44 −4 ) than postural instability gait difficulty (PIGD) 6.06 −2 ). The rs2253820 PIGD 5.42 TD 4.2 Haplotype analysis also showed were PD. Imputation identified some which affect gene expression through altering transcription factor binding site. In summary, our findings suggest genetic polymorphisms genes, as well disruptions, pathogenesis.
