The infection by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is associated not only with the development of disease but also long-term symptoms or post-acute sequelae SARS-CoV-2 (PASC). Multiple lines evidence support that some viral antigens and RNA can persist for up to 15 months in multiple organs body, often after apparent clearance from upper system, possibly leading persistence symptoms. Activation immune system observed a prolonged time, providing indirect elements infection. In gastrointestinal tract, could stimulate shaping local microbiota potential systemic effects. All these interactions need be investigated, taking into account predisposing factors, multiplicity pathogenic mechanisms, stratifying populations vulnerable individuals, particularly women, children, immunocompromised where may present additional challenges.

To investigate the effectiveness of platelet-rich plasma (PRP) injection into olfactory clefts coronavirus disease 2019 (COVID-19) patients with persistent anosmia, hyposmia, and parosmia. Controlled study. Multicenter From March 2022 to July 2024, 514 COVID-19 undergoing PRP training versus (controls) for a dysfunction (OD) were consecutively recruited from two medical centers. Subjects evaluated pre- 3 months post-injection disorder questionnaire (ODQ) threshold, discrimination, identification (TDI) test. Olfactory outcomes compared between controls considering type OD (anosmia, parosmia). The group was composed 416 anosmia (N = 111; 26.7%), hyposmia 160; 38.5%), parosmia 145; 34.9%). baseline ODQ significantly higher than those other groups (P .001). increase in TDI subscores total score all subgroups 0.001). reduced post-PRP Patients showed greater reduction scores control group. 3-month negatively influenced by age (rs -0.263; P 0.001) duration -0.213; experienced stronger impact on quality life (QoL), QoL improvement following injections groups.

The pathophysiological mechanisms of the post-acute sequelae COVID-19 (PASC) remain unclear. Sex differences not only exist in disease severity acute SARS-CoV-2 infection but also risk suffering from PASC. Women have a higher PASC and longer time to resolution than men. To explore possible immune among non-elderly females, we mined single-cell transcriptome data peripheral blood samples female patients with infection, together age- gender-matched non-PASC healthy controls available Gene Expression Omnibus database. By comparing differences, found that CD14+ monocyte subset characterized by expression signal transducers activators transcription 2 (STAT2) (CD14+STAT2high) was notably increased compared individuals. transcriptional factor (TF) activity analysis revealed STAT2 IRF9 were key TFs determining function CD14+STAT2high monocytes. are exclusively involving type I III interferon (IFN) signaling pathways, resulting uncontrolled IFN-I activation interferonopathy. Furthermore, common interferon-stimulated genes (ISGs) has been identified most subsets patients, including IFI6, IFITM3, IFI44L, IFI44, EPSTI1, ISG15, MX1. This study reveals featured associated activation, which is indicative interferonopathy
