They commonly used as biopesticides for pest control due to their significant practical value. Abstract BACKGROUND Species that experience outbreaks and those display density‐dependent phase polymorphism demonstrate prophylaxis (DDP) by increasing their immune investment in response to densities. Despite this phenomenon, the mechanisms of DDP remain largely unexplored. RESULTS Here, we showed Spodoptera litura exhibited heightened cuticular melanization enhanced responses when reared at higher population density. Transcriptomic analysis identified differentially expressed genes (DEGs) associated with responses, nutritional metabolism, synthesis cuticle, revealing molecular underpinnings plasticity larval cuticles. Gregarious S. larvae significant up‐regulation immune‐related genes, particularly Toll deficiency (IMD) signaling pathways tyrosine suggesting a strategic enhancement defenses. Concurrently, reduced lipid metabolism was observed cuticle gregarious larvae, suppressed expression key fatty acid synthesis, leading decrease integument triglyceride content. The Gti1/Pac2 protein family, which is highly conserved across fungi, pivotal in processes such as fungal development, spore formation, export, toxin production, and virulence. Despite its importance, the precise functions of Pac2 within entomopathogenic fungi have yet to be fully understood. In our study, MaPac2 gene from M. acridum was identified, were explored. Social immunity-mediated sanitation behaviours occur in insects when microbially killed corpses are removed and/or dismembered by healthy nestmates. However, little is known concerning the chemical signals or receptor proteins that mediate these responses. SiFABP5 bound all three dismemberment-related compounds, whereas interactions between SiApoLp-III narrowed to acid. Pear psylla (Cacopsylla chinensis), a major pear tree pest widely distributed in China, is increasingly affecting the productivity of orchards. This species exhibits seasonal polyphenism with two distinct forms, namely, summer form and winter form. Through topically applying Beauveria bassiana conidial suspensions to abdominal cuticle C. chinensis, we demonstrated that entomopathogenic fungus B. significant yet phenotypically divergent virulence against these forms. Using PacBio SMRT sequencing Illumina RNA-seq, analyzed transcriptomic changes post-infection, revealing form-specific immune responses, 18,232 5027 differentially expressed genes identified summer- winter-form psylla, respectively, total 3715 DEGs shared between phenotypes. In summer-form individuals, infection disrupted oxidative phosphorylation downregulated recognition genes, cellular immune-related signaling along upregulation inhibitor serpin, indicating immunosuppression. Conversely, glycolytic rate-limiting enzymes were upregulated after infection, suggesting system normally responds supports efficient defense through metabolic reprogramming fuel energy-demanding defenses. These findings advance our understanding chinensis/B. interactions, providing basis for elucidating regulation seasonally polymorphic insects.
