Liver diseases are a significant global health issue, characterized by elevated levels of disorder and death. The substantial impact ageing on liver their prognosis is evident. Abstract Aging is characterized by a decline in physiological functions and an increased susceptibility to age-related diseases. This study investigates the therapeutic potential of mesenchymal stem cells (MSCs) pyrroloquinoline quinone (PQQ), individually combination, counteract aging-related declines, with specific focus on their modulation AMP-activated protein kinase (AMPK) pathway, key regulator cellular energy homeostasis stress response. was induced thirty-seven female rats using D-galactose, simulating metabolic imbalances oxidative characteristic aging. The experimental groups included controls, aged without treatment, treated MSCs, PQQ, or combined MSC-PQQ regimen. MSC homing analyses Behavioral assessments, assays, gene expression profiling, histopathological evaluations were conducted provide multidimensional view treatment efficacy. confirmed successful tissue localization repair, underscoring regenerative capacity MSCs. Remarkably, therapy (APQQST) markedly improved anxiety-related behaviors, evidenced rearing grooming activities (p < 0.01). Oxidative biomarkers supported these findings; exhibited significantly reduced malondialdehyde (MDA) levels elevated antioxidant defenses, including glutathione (GSH) peroxidase (GPX) Gene analysis highlighted beneficial upregulation genes such as LKB1, PFKFB3, TSC2, HMGR, crucial for response, combination showing most pronounced effects. Furthermore, assessments underscored significant liver recovery groups, particularly (APQQST), minimal vacuolar degeneration restored hepatic architecture These findings highlight synergistic effects MSCs PQQ mitigating behavioral, molecular, aspects aging, promising agents promoting healthy aging offering foundation future translational research clinical applications.
