Acute kidney injury (AKI) is a significant issue in public health, displaying high occurrence rate and mortality rate. Ferroptosis, form of programmed cell death (PCD), characterized by iron accumulation intensified lipid peroxidation. Recent studies have demonstrated the pivotal significance ferroptosis AKI caused diverse stimuli, including ischemia-reperfusion (IRI), sepsis toxins. Autophagy, multistep process that targets damaged organelles macromolecules for degradation recycling, also plays an essential role AKI. Previous research has autophagy deletion proximal tubules could aggravate tubular renal function loss, indicating protective Consequently, finding ways to stimulate become crucial therapeutic strategy. The recent discovery selective influencing identified new highlighted importance understanding cross-talk between ferroptosis. This study aims provide overview signaling pathways involved autophagy, focusing on mechanisms functions autophagy-dependent We hope establish foundation future investigations into interaction as well other diseases.

Liver fibrosis accompanies the progression of chronic liver diseases independent etiologies, such as hepatitis viral infection, alcohol consumption, and metabolic-associated fatty disease. It is commonly associated with injury, inflammation, cell death. characterized by abnormal accumulation extracellular matrix components that are expressed myofibroblasts collagens alpha-smooth actin proteins. Activated hepatic stellate cells contribute to major population myofibroblasts. Many treatments for have been investigated in clinical trials, including dietary supplementation (e.g., vitamin C), biological treatment simtuzumab), drug pegbelfermin natural herbs), genetic regulation non-coding RNAs), transplantation stem hematopoietic cells). However, none these has approved Food Drug Administration. The efficacy can be evaluated histological staining methods, imaging serum biomarkers, well scoring systems, fibrosis-4 index, aspartate aminotransferase platelet ratio, non-alcoholic disease score. Furthermore, reverse slowly frequently impossible advanced or cirrhosis. To avoid life-threatening stage fibrosis, anti-fibrotic treatments, especially combined behavior prevention, treatment, drugs herb medicines, needed. This review summarizes past studies current future fibrosis.

Excess free iron is a substrate for the formation of reactive oxygen species (ROS), thereby augmenting oxidative stress. Oxidative stress well-established cause organ damage in liver, main site storage. Ferroptosis, an iron-dependent mechanism regulated cell death, has recently been gaining attention development and progression liver disease. We therefore summarize mechanisms metabolism, its close connection to ferroptosis, particular relevance disease metabolic-dysfunction-associated fatty potential targets therapy from clinical perspective.

Colorectal cancer (CRC) is a common malignant tumor of the digestive system, and pathogenic mechanism still unclear, mostly related to genetics, immunity, inflammation, abnormal activation tumor-related signaling pathways. MAPK belongs Ser/Thr kinase family, which plays an important role in complex cellular programs such as regulation cell proliferation, differentiation, apoptosis, angiogenesis, metastasis. Increasing evidence supports that highly correlated with risk CRC. Targeting may be therapeutic strategy, natural products show great potential regulating MAPK-related proteins. In this paper, we searched PubMed, Web Science CNKI databases keywords "colorectal cancer, products, pathway, ERK, P38, JNK" for relevant studies last 14 years from 2010 2024. This work retrieved 47 studies, aiming provide new strategies CRC patients lay foundation drug development.

Abstract: Ferroptosis, a unique form of programmed cell death, is initiated by an excess iron accumulation and lipid peroxidation-induced damage. There growing body evidence indicating that ferroptosis plays critical role in the advancement tumors. The increased metabolic activity higher levels tumor cells make them particularly vulnerable to ferroptosis. As result, targeted induction becoming increasingly promising approach for cancer treatment. This review offers overview regulatory mechanisms ferroptosis, delves into mechanism action traditional small molecule inducers their effects on various In addition, latest progress inducing using new means such as proteolysis-targeting chimeras (PROTACs), photodynamic therapy (PDT), sonodynamic (SDT) nanomaterials summarized. Finally, this discusses challenges opportunities development ferroptosis-inducing agents, focusing discovering targets, improving selectivity, reducing toxic side effects. BACKGROUND Liver fibrosis is a compensatory response during the tissue repair process in chronic liver injury, and finally leads to cirrhosis or even hepatocellular carcinoma. The pathogenesis of hepatic associated with progressive accumulation activated stellate cells (HSCs), which can transdifferentiate into myofibroblasts produce an excess extracellular matrix (ECM). Myofibroblasts are main source excessive ECM responsible for fibrosis. Therefore, (aHSCs), principal producing injured liver, promising therapeutic target treatment AIM To explore effect taurine on aHSC proliferation mechanisms involved. METHODS Human HSCs (LX-2) were randomly divided five groups: Normal control group, platelet-derived growth factor-BB (PDGF-BB) (20 ng/mL) treated low, medium, high dosage (10 mmol/L, 50 100 respectively) PDGF-BB group. Cell Counting Kit-8 method was performed evaluate viability aHSCs. Enzyme-linked immunosorbent assay used estimate levels reactive oxygen species (ROS), malondialdehyde, glutathione, iron concentration. Transmission electron microscopy applied observe autophagosomes ferroptosis features Quantitative real-time polymerase chain reaction Western blot analysis detect expression α-SMA, Collagen I, Fibronectin 1, LC3B, ATG5, Beclin PTGS2, SLC7A11, p62. RESULTS Taurine promoted death aHSCs reduced deposition ECM. Treatment could alleviate autophagy inhibit their activation, by decreasing autophagosome formation, downregulating LC3B 1 protein expression, upregulating p62 expression. Meanwhile, triggered ferritinophagy eliminate characterized overload, lipid ROS accumulation, glutathione depletion, peroxidation. Furthermore, bioinformatics demonstrated that had direct targeting nuclear receptor coactivator 4, exhibiting best average binding affinity -20.99 kcal/mol.
