Chronic neuropathic pain and comorbid depression syndrome (CDS) is a major worldwide health problem that affects the quality of life patients imposes tremendous socioeconomic burden. More than half with chronic also suffer from moderate or severe depression. Due to complex pathogenesis CDS, there are no effective therapeutic drugs available. The lack research on neural circuit mechanisms CDS limits development treatments. purpose this article provide an overview various circuits involved in CDS. Notably, activating some can alleviate and/or depression, while other exacerbate these conditions. Moreover, we discuss current emerging pharmacotherapies for such as ketamine. Diffuse axonal injury (DAI) disrupts the integrity of white matter microstructure and affects brain functional connectivity, resulting in persistent cognitive, behavioral affective deficits. Mounting evidence suggests that altered cortical-subcortical connectivity is a major contributor to cognitive dysfunction. The striatum particularly vulnerable DAI, but has received less attention. This study aimed investigate alteration patterns striatal subdivision connectivity. Twenty-six patients with DAI 27 healthy controls underwent resting-state fMRI scans on 3.0 T scanner. We assessed using seed-based analysis DAI. Furthermore, partial correlation was used measure its clinical association. Compared controls, showed decreased between right inferior ventral frontal gyrus, as well parietal lobule, left superior bilateral cerebellar posterior lobe, dorsal caudal putamen anterior cingulate lobule. Our study aims to explore the differences in functional connectivity nucleus accumbens (NAc) between patients with melancholic depression and non-melancholic (NMD) their relation depression's pathogenesis. We recruited 60 depression, 58 NMD, 80 healthy controls, all matched for gender, age, education. Functional analysis focused on bilateral NAc as region of interest, comparing it whole brain correlating significant clinical scores. Melancholic showed reduced left anterior regions, right temporal frontal areas, compared controls. In contrast, NMD displayed only posterior cingulate cortex. also exhibited increased middle gyrus, unlike patients. The findings suggest that exhibit unique patterns, particularly default mode network prefrontal suggesting atypical reward-circuitry interactions. NAc's connection gyrus may distinguish from NMD.

Objective Depression is a prevalent mental disorder characterized by persistent negative mood and loss of pleasure. Although there are various treatment modalities available for depression, the rates response remission remain low. Xiaoyaosan (XYS), traditional Chinese herbal formula with long history use in treating has shown promising effects. However, underlying mechanism its therapeutic action remains elusive. Anhedonia is an enduring symptom of subthreshold depression (StD) and predict later onset major depressive disorder (MDD). Brain structural covariance describes the inter-regional distribution morphological changes compared to healthy controls (HC) reflects brain maturation disease progression. We investigated neural correlates anhedonia from covariance. T1-weighted magnetic resonance images were acquired 79 young adults (26 StD, 30 MDD, 23 HC).
