Abstract Background Oral diseases with high prevalence worldwide are recognized as severe health problems. Probiotics used to prevent oral diseases, including dental caries, malodor, periodontitis, and subgingival plaque. In this study, we aimed confirm the antibacterial effect of probiotics on pathogens assess their characterization safety probiotics. Methods The effects Lacticaseibacillus rhamnosus MG4706, paracasei MG4715, Limosilactobacillus reuteri MG4722 growth biofilm formation Streptococcus mutans , Aggregatibacter actinomycetemcomitans Porphyromonas gingivalis were evaluated. We also investigated production substances (H 2 O reuterin) by these strains ability adhere epithelial cells. L. was verified through whole-genome sequencing analysis antibiotic susceptibility, lactate dehydrogenase activity, hemolytic bile acid hydrolase activity. reuterin biosynthesis genes identified using genomic analysis. Results significantly inhibited S . A. P. suppressed addition, it showed considerable adhesion produced H substances, confirmed presence encoding compounds reuterin, reuteran, reutericyclin. no hemolysis, salt resistance or toxicity HT-29 cells, antibiotic-resistance identified. Conclusion demonstrated producing adhering These results suggest that could be an effective probiotic for health.

The human oral cavity contains a diversity of microbial habitats that have been adopted and adapted to as homeland by an amazingly heterogeneous population microorganisms collectively referred the microbiota. These microbes generally co-habit in harmonious homeostasis. However, under conditions imposed stress, with changes host’s physiology or nutritional status, response foreign antimicrobial incursions, some components “microbiome” (viz. situ microbiota) may enter dysbiotic state. This microbiome dysbiosis can manifest variety guises including streptococcal sore throats, dental caries, thrush, halitosis periodontal disease. Most strategies currently available for management treatment diseases focus on repetitive “broad sweep” short-term culling microbe populations, hopefully perceived principal pathogens. Both physical chemical techniques are used. application more focused approaches harnessing elimination key pathogens is now feasible through use probiotic strains naturally colonization also equipped produce anti-competitor molecules such bacteriocins bacteriocin-like inhibitory substances (viz BLIS). Some these probiotics capable suppressing proliferation recognized mouth, thereby assisting restoration BLIS K12 M18, progenitors BLIS-producing probiotics, members commensal species Streptococcus salivarius. More recently however, number other non-streptococcal candidate promoted. What becoming increasingly apparent future applications will probably extend well beyond attempted limitation direct pathological consequences encompass plethora systemic disorders host. background evolving prospects beneficial modulation via S. salivarius comprises present review.

Oral health is critical for total body and well-being; however, little improvement in oral status has occurred the U.S. over past 20 years. Tooth decay gum disease remain highly prevalent, with more than 90% 50% of adults suffering from these conditions, respectively. To combat this lack improvement, alternative approaches to dental care are now being suggested. One such therapy probiotics care. In cavity, probiotic strains have been shown reduce levels pathogens, inhibit formation caries, bacteria that cause halitosis. However, as market expands, many products contain bacterial species no documented benefits leading confusion mistrust among consumers clinicians. This enhanced by regulatory which puts onus safety efficacy on manufacturer rather a central body. The overarching goal review provide clinicians evidence supporting (or refuting) marketed sale United States. includes defining what constitutes an product strain level analysis candidate genera Streptococcus, Lactobacillus, Bifidobacterium, Bacillus. Additionally, prebiotics postbiotics will be discussed. Finally, set considerations provided empower decision making. Together, improve understanding US professionals consumers.

To identify which methods have been used to deliver adjunctive probiotics during the non-surgical management of periodontitis in clinical trials. This review also investigates and at what dose, along with periodontal treatment outcomes achieved. Conducted accordance Joanna Briggs Institute methodology for scoping reviews. MEDLINE, Embase, Web Science, Scopus were searched on 02/02/2024 from inception no date limits applied. Clinical trials investigating use as an therapy humans. Out 4769 studies 66 met inclusion criteria. Over 30 different identified 18 delivery varying dosages duration. 48 included reported a beneficial effect when probiotic was used, 14 difference, 3 found comparable antibiotics, 1 study better outcome control group. The its dosage, method, duration application, measures differ across making it difficult draw conclusions their efficacy. highlights need further research establish uniform protocol most effective bacteria. Despite majority indicating potential benefit periodontitis, high level heterogeneity between interventions they currently cannot be recommended practice.

Abstract Objective This study investigates the regenerative and protective effects of postbiotics (cell-free supernatant) derived from Lactiplantibacillus plantarum EIR/IF-1 strain on human periodontal ligament mesenchymal stromal cells (hPDL-MSCs). Materials methods hPDL-MSCs were isolated tissues (PDL) wisdom teeth using enzymatic digestion subsequently characterized through immunophenotyping. The effect viability was assessed MTT assay flow cytometry, while their impact cell migration evaluated via scratch assay. Anti-inflammatory investigated lipopolysaccharide (LPS, Porphyromonas gingivalis )-stimulated Enzyme-Linked Immunosorbent Assay (ELISA). Additionally, antioxidant analyzed in hydrogen peroxide (H₂O₂)-induced by measuring reactive oxygen species (ROS) levels cytometry. expression collagen type I ( COL1A1 ) gene further quantitative reverse transcription PCR immunofluorescence staining. Results Treatment with (250 µg/mL) significantly increased capability hPDL-MSCs, enhancing production for PDL repair. 24 h resulted a 54.53 ± 2.01% reduction intracellular ROS compared to untreated H 2 O -induced hPDL-MSCs. Furthermore, decreased pro-inflammatory cytokines (IL-8, IL-6, IL-1β), anti-inflammatory cytokine IL-10 (2.67-fold) LPS-stimulated Conclusion Our findings indicate that exhibit biological activity throughout all stages healing process, beginning modulation inflammatory response LPS stimulation, followed promotion migration, proliferation, synthesis. Given unmet need safe adjuvant therapeutic approaches promote comprehensive regeneration diseases, this presents as promising candidate. Clinical relevance Postbiotics could be integrated into therapies novel bioactive material improve outcomes defects both controlling inflammation stimulating tissue repair processes.
