The coronavirus disease 2019 (COVID-19) pandemic has significantly affected older adults. Identifying host COVID-19 susceptibility genes in elderly populations remains a challenge. Here, we aimed to identify genetic factors influencing the severe acute respiratory syndrome 2 (SARS-CoV-2) infection. We genotyped 12 single-nucleotide polymorphisms (SNPs) previously associated with innate immune response total of 97 (age > 65 years) residents three long-term care facilities located Barcelona, Spain. Individuals were PCR-tested during SARS-CoV-2 outbreaks between September and November 2020. PCR tests revealed infections 81 residents. Importantly, 16 uninfected remained seronegative until vaccination (January February 2021). After adjusting for sex age, found that two SNPs infection susceptibility—MMS19 nucleotide excision repair protein homolog (MMS19)/rs2236575 (p = 0.029) interferon-induced helicase C domain-containing 1 (IFIH1)/rs1990760 0.034). No association was 10 additional SNPs, which included 4 on chromosome gene encoding oligoadenylate synthetase (OAS). Our results indicate MMS19/rs2236575_A IFIH1/rs1990760_TC variants resistance cohort institutionalized seniors.

The evolution of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) continues to challenge the efficacy vaccination efforts against disease 2019 (COVID-19). Omicron XBB lineage SARS-CoV-2 has presented dramatic evasion neutralizing antibodies stimulated by mRNA and COVID-19 convalescence. XBB.1.16, characterized two mutations relative dominating variant XBB.1.5, i.e., E180V K478R, been on rise globally. In this study, we compare immune escape XBB.1.16 with alongside ancestral variants D614G, BA.2, BA.4/5. We demonstrate that is strongly evasive, extent comparable XBB.1.5 in bivalent-vaccinated healthcare worker sera, 3-dose-vaccinated BA.4/5-wave convalescent sera. Commencing in December 2019 with the emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), three years disease (COVID-19) pandemic have transpired. Genomic surveillance of positive SARS-CoV-2 samples is important to monitor the genetic changes occurring in virus, this was enhanced after WHO designation XBB.1.16 as a variant under monitoring March 2023. From 5th February till May 6, 2023 all were monitored for changes. A total 1757 having Ct value <25 (for E and ORF gene) from different districts Rajasthan processed Next Generation Sequencing (NGS). The FASTA files obtained on sequencing used lineage determination using Nextclade phylogenetic tree construction. identification done 1624 samples. predominant 1413 (87.0%) cases while rest other XBB (207, 12.74%) lineages (4, 0.2%). Of cases, 57.47% males 42.53% females. Majority (66.53%) belonged 19–59 year age. 84.15% infected first time. Hospitalization required only 2.2% death reported 5 (0.35%) patients. Most symptomatic commonest symptoms fever, cough rhinorrhea. Co-morbidities present 414 (29.3%) cases. Enhanced genomic helped rapidly identify spread Rajasthan.
