SUMMARY Sexual reproduction in malaria parasites is essential for their transmission to mosquitoes. It also offers a divergent eukaryote model understand the evolution of sex. Through panel genetic screens, where each sex Plasmodium berghei was mutagenised separately with barcoded vectors, we identify 401 and transmission-related gene functions define roles hundreds unstudied fertility genes as putative targets blocking interventions. The functional data provide deeper understanding female metabolic reprogramming, meiosis axoneme. We protein complex SUN domain protein, SUN1, moonlighting allantoicase, ALLC1, that male by linking microtubule organising centre nuclear envelope enabling mitotic spindle formation during gametogenesis. Both proteins have orthologs mouse testis, point an ancient role atypical fertility. Altogether, our unbiased picture molecular processes underpin parasite but highlight ancestral aspects evolved close last eukaryotic common ancestor.

Abstract Microsporidia are parasites with the smallest known eukaryotic genomes. The extent of protein loss in these organisms has been well documented, but much less is about how compaction microsporidia proteins affects their function. Taking a comparative genomic approach, we identified orthologs budding yeast and show that enriched for essential genes. We median 21% shorter than its counterpart although extensive occurred after divergence microsporidia, reduced sizes were already present microsporidian relatives. through domain lengths, diminished linker loss, having lost domains yeast. On average, 34% have C-terminal residues function yeast, including 13 per genome. also found display distinct phylogenetic patterns losses occurring clade-specific manner. To investigate conservation function, used complementation assays to test from several species relative Rozella allomycis . These experiments reveal most cannot complement orthologs, ability three-fold observed R. do not more length orthologs. Altogether, our results demonstrate drastic reduction reductions resulted functional fungal ancestors.
