Abstract Organoid models have advanced our understanding of adult stem/progenitor cell dynamics and function. However, uncovering the regulatory mechanisms scarce often quiescent stem cells in organs like salivary glands remains challenging. Using single-cell RNA sequencing bulk ATAC RNA-sequencing analysis, we conducted in-depth profiling cellular populations key signaling pathways characterizing a mouse submandibular gland organoid (mSGO) model at different temporal stages response to radiation damage. We identified Sox9- Itgb1- expressing as most primitive uncover novel stemness migratory roles for Cd44 -expressing cells. Moreover, revealed that Notch is essential maintaining self-renewal migration potential these post-irradiation. Extending findings patient-derived mSGOs, well murine mammary thyroid organoids, confirmed conserved regulator function under migrative regenerative conditions.

The use of charged particle radiotherapy is currently increasing, but combination therapy with DNA repair inhibitors remains to be exploited in the clinic. high-linear energy transfer (LET) radiation delivered by particles causes clustered damage, which particularly effective destroying cancer cells. Whether damage response this type different from that elicited low-LET radiation, and if how it can targeted increase treatment efficacy, not fully understood. Although several preclinical studies have reported radiosensitizing effects when proton or carbon ion irradiation combined of, e.g., PARP, ATR, ATM, DNA-PKcs, further exploration required determine most treatments.
