Animal tissues comprise diverse cell types. However, the mechanisms controlling number of each type within tissue compartments remain poorly understood. Here, we report that different types utilize distinct strategies to control population numbers. Proliferation fibroblasts, stromal cells important for integrity, is limited by space availability. In contrast, proliferation macrophages, innate immune involved in defense, repair, and homeostasis, constrained growth factor Examination density-dependent gene expression fibroblasts revealed Hippo TGF-β target genes are both regulated density. Cancer is a multistep disease driven by the activation of specific oncogenic pathways concomitantly with loss function tumor suppressor genes that act as sentinels to control physiological growth. Cancer, a complex yet common disease, is caused by uncontrolled cell division and abnormal growth due to variety of gene mutations. Seeking effective treatments for cancer major research focus as the incidence on rise drug resistance existing anti-cancer drugs concern. Natural products have potential yield unique molecules combinations substances that may be against with relatively low toxicity/better side effect profile compared standard anticancer therapy. Drug discovery work natural has demonstrated compounds display wide range biological activities correlating effects. In this review, we discuss formononetin (C16H12O4), which originates mainly from red clovers Chinese herb Astragalus membranaceus. The compound comes class 7-hydroisoflavones substitution methoxy group at position 4. Formononetin elicits antitumourigenic properties in vitro vivo modulating numerous signaling pathways induce apoptosis (by intrinsic pathway involving Bax, Bcl-2 caspase-3 proteins), cycle arrest regulating mediators like cyclin A, B1 D1), suppress proliferation STAT activation, PI3K/AKT MAPK pathway) inhibit invasion factors VEGF FGF2, MMP-2 MMP-9 proteins). Co-treatment other chemotherapy such bortezomib, LY2940002, U0126, sunitinib, epirubicin, doxorubicin, temozolomide metformin enhance both respective through synergistic effect. Ageing leads to dramatic changes in the physiology of many different tissues resulting a spectrum pathology. Nonetheless, lines evidence suggest that ageing is driven by highly conserved cell intrinsic processes, and set unifying hallmarks has been defined. Here, we survey reports age-linked basal gene expression across eukaryotes from yeast human identify six cellular ageing: downregulation genes encoding mitochondrial proteins; protein synthesis machinery; dysregulation immune system genes; reduced growth factor signalling; constitutive responses stress DNA damage; mRNA processing.
