Neuropeptidomic data were collected on the mosquito Ae. aegypti, which is considered most tractable species for physiological and endocrine studies. The solely obtained by direct mass spectrometric profiling, including tandem fragmentation, of selected tissues from single specimens, yielded a largely complete accounting putative bioactive neuropeptides; truncated neuropeptides with low abundance not counted as mature peptides. Differential processing within CNS was detected CAPA-precursor, differential post-translational (pyroglutamate formation) AST-C CAPA-PVK-2. For first time in insects, we succeeded profiling midgut tissue comprehensive immediate overview peptides involved system gut. Head earlier identified abundant RFamides any part or midgut. This study provides framework future investigations endocrinology neurobiology. Given high sequence similarity neuropeptide precursors other medically important mosquitoes, conclusions regarding peptidome aegypti likely are applicable to these mosquitoes.

The synthesis of vitellogenin and its uptake by maturing oocytes during egg maturation are essential for successful female reproduction. These events regulated the juvenile hormones ecdysteroids nutritional signaling pathway neuropeptides. Juvenile act as gonadotropins, regulating vitellogenesis in most insects, but control this process Diptera some Hymenoptera Lepidoptera. complex crosstalk between hormones, ecdysteroids, pathways differs distinctly depending on reproductive strategies adopted various insects. Molecular studies within past decade have revealed much about relationships among, role of, these with respect to regulation insect Here, we review signaling, along that microRNAs, reproduction at molecular level.

Neuropeptides are signaling molecules that commonly act via G protein-coupled receptors (GPCRs) and generated in neurons by proneuropeptide (pNP) cleavage. Present both cnidarians bilaterians, neuropeptides represent an ancient widespread mode of neuronal communication. Due to the inherent difficulties analyzing highly diverse repetitive pNPs, relationships among different families often elusive. Using similarity-based clustering sensitive similarity searches, I obtained a global view metazoan pNP diversity evolution. Clustering revealed large diffuse network sequences connected significant sequence encompassing one-quarter all families. pNPs belonging this cluster were also identified early-branching neuronless animal Trichoplax adhaerens . neuropeptide GPCRs several orthology groups allowed reconstruction phyletic distribution receptor GPCR closely paralleled indicating extensive conservation long-term coevolution receptor–ligand pairs. Our recent studies identified juvenile hormone (JH) and nutrition as the two key signals that regulate vitellogenin (Vg) gene expression in red flour beetle, Tribolium castaneum. Juvenile regulation of Vg synthesis has been known for a long time several insects, but mechanism JH action is not known. Experiments were conducted to determine these expression. Injection bovine insulin or FOXO double-stranded RNA into previtellogenic, starved, JH-deficient female adults increased mRNA protein levels, thereby implicating pivotal role insulin-like peptide signaling possible cross-talk between pathways. Reduction its by RNAi-mediated silencing genes coding acid methyltransferase methoprene-tolerant decreased peptides (ILPs) influenced subcellular localization, resulting down-regulation Furthermore, application previtellogenic beetles induced ILP2 ILP3, expressed baculovirus system binds response element present promoter. These data suggest functions through pathway
